Objective: To study the influence and pathway cf invasion and metastasis between mesenchymal stem cells (Mesenchymal Stem Cell, MSC) and colon cancer in vivo, and to provide the theoretical basis for the clinical applications of mesenchymal stem cells and new access to treatment of colon cancer.Methods: The model of colon cancer will be made to plante the cell of colon cancer into nude mice, then the nude mice will be divided into test group and control group, the nude mice in test group will receive mesenchymal stem cells by intravenous injection and the control group will be given placebo. After 15 days tumor block will be removed and measured by the size and weight; then the tumor block will be made into paraffin mass, then determinate the expression of focal adhesion kinase (focal adhesion kinase, FAK) of the two groups with the methods of SP and RT-PCR. All the data will be processed with the statistical software of SPSS13.0 for statistical analysis. The data of mRNA will be indicated with x±s,and the comparison between the two groups will be processed using t test; then the comparison of results of immunohistochemistry is to use X2 test, using two-sided test, and is considered statistically significant with P <0.05.Results: (1) The volume and weight of tumor in nude mice of test group (0. 3428±0. 0874) (0. 4275±0. 0628) were less than the control group (0. 5366±0. 0531) (0. 5923±0. 0611), the difference was statistically significant (P <0.01), that mesenchymal stem cells in vivo can inhibit the growth of colon cancer cells. (2) The positive rate of FAK in the test group (70%) was lower than the control group(90%), the difference was statistically significant (P <0.05), that mesenchymal stem cells in vivo can inhibit invasion and metastasis of colon cancer cells . (3)Relative expression level of FAKmRNA in the test group (0.32±0.19) was lower than the control group (0.66±0.28), the difference was statistically significant (P <0.05), that mesenchymal stem cells can suppress invasion and metastasis of colon cancer cells in vivo.Conclusion: The mesenchymal stem cells can suppress the growth and invasion and metastasis of colon cancer cells in nude mice by inhibiting the pathway of FAK, mesenchymal stem cells have a broad prospect in clinical research and cancer treatment in future.
