Objective: Influence of Cyclophosphamide on Regulatory T Cells of BALB/c for breast cancer by tumors and transfer lymph node in mice. To investigate the influence of the chemotherapy drugs cyclophosphamide (CTX) on regulatory T Cells of BALB/c for breast cancer by tumors and transfer lymph node in mice, discuss chemotherapy drugs for tumor microenvironment and transferring tumor microenvironment immunosup- pressive cells in the role, to establish Cellular immune therapy of new method based on chemotherapy intervention tumor microenvironment immune therapy provide theoretical support and experimental data.Methods: BALB/c mouse tumor model was establishing by inoculating 4T1 cells.32 mice were randomly divided into two groups;16 mice in each group; control group were injected with PBS around the tumor treatment group were injected with cyclophosphamide around the tumor; Determined for CD4+CD25+Foxp3+ regulatory T cells content in tumor microenvironment and transferring tumor microenvironment in 2 groups of mice by flow cytometry(FCM)Results:1 Statistics based on the results of FCM, In control group that PBS were given in the tumor tissue before and after 4 days, 8 days, 12 days the relative number of CD4+CD25+Foxp3+regulatory Tcells are(9.975±0.275)%,(18.775±1.289) %,(21.950±3.718) %,(25.025±1.743) %. The treatment group given Cyclophosphamide in the tumor tissue before and after 4 days, 8 days, 12 days the relative number of CD4 + CD25 + Foxp3 regulatory T cells are(9.850±0.264)%,(7.075±1.233)%,(6.225±1.030)%,(8.000±0.424)%, there were significant differences in the two groups of comparisons.(p<0.05). 2 According to statistical by FCM, Metastatic lymph nodes in the control group given PBS before and after the first 4 days, 8 days, 12 days the relative number of CD4 + CD25 + Foxp3regulatory T cells are ( 2.150±0.264)% (7.675±0.275)%,(13.100±0.787) %,(15.100±0.734) %. Drug treatment group before and after administration in 4 days, 8 days, 12 days the relative number of CD4 + CD25 + Foxp3 regulatory T cells are (2.150±0.264)%,(4.150±0.732)%,(2.900±0.588)%,(4.550±0.591)%.There were significant differences in the two groups of comparisons. (p<0.05)Conclusions:1 Cyclophosphamide can effectively reduce the CD4 + CD25 + Foxp3 +levels of regulatory T cells in the microenvironment tumor tissue of breast cancer in mice2 Cyclophosphamide can effectively reduce the CD4 + CD25 + Foxp3+ levels of regulatory T cells in transfer lymph node of breast cancer in mice The results of this study hints Cyclophosphamide as a class of conventional chemotherapy drugs besides can effectively reduce the level of CD4 + CD25 + Foxp3 regulatory T cells in primary tumor local organizations , still can effectively reduce the level of CD4 + CD25 + Foxp3 regulatory T cells in lymph node metastasis. So as to interfere with the tumor microenviro- nment of cyclophosphamide on the basis of adoptive immunotherapy to provide a new basis and reference.
