Objective To observe the efficacy and discuss the mechanism of Dragon's Blood in the treatment of active ulcerative colitis and clinical experimental investigation.Methods1. Experimental research methods:60 SD rats were randomly divided into 5 groups-blank group, control group and the low, medium and high-concentration Dragon's Blood group。The blank group was given nothing, the control group, low, medium and high concentration of Dragon's Blood group were given 3% DSS solution free drink 7 days, resulting in acute UC model。After the modeling, every day the blank group and control group given normal saline 4ml/only by gavage, and the other groups were given oral Dragon's Blood treated with different concentrations of the compound 4ml/only by gavage were fed for 7 days. Rats were killed, the model DAI, HI rate, measured the expression of plasma PAF and PF4 and observed the pathology of rectum and colon.2. Clinical research methods:68 patients with blood stasis type of Mild-to-moderate UC were randomly divided into treatment group and control group, All patients oral SASP tablets, lg tid treatmented group treated with Dragon's Blood enema at bedtime and controlled group treated with Hydrocortisone sodium succinate enema at bedtime.Both groups were treated for one months and then six month follow-ups. Evaluated the clinical efficacy, improvement of TCM symptoms and adverse reactions.Results1. Experimental research results:The DAI, HI and the expression of plasma PAF and PF4 of blank group,and low, medium and high concentration of Dragon's Blood group compared with the control group was statistically significant (P<0.05); control group,and low, medium and high concentration of Dragon's Blood group compared with the blank group was statistically significant (P<0.05); Low, medium and high concentration of the Dragon's Blood group was not significant between the two groups (P> 0.05)。Although the low, medium and high concentration of the Dragon's Blood groups was not significant between the two groups, but the data in the table and PAF, PF4 means plot shows from low to high base a downward trend, especially in low and high concentrations of groups more apparent.2. Clinical research results:The total effective rate of the treatmented group was 93.9%, and the controlled group was 87.5%,no significant difference (P>0.05)between the two groups;The improvement of TCM symptoms effective rate of the treatmented group was 84.8%, and the controlled group was 46.7%,was Statistically significant(P<0.05) between the two groups,Adverse events compared was statistically significant(P<0.05) between the two groups,the treatmented group was significantly better than the control group.Conclusion To a certain extent,the Dragon's Blood can inhibit platelet activation, reduced expression of plasma PAF and PF4,this effect is very significant. This is one of the mechanisms for treatmented UC. The Dragon's Blood can improve blood stasis for mild-moderate UC patients,and low rate of adverse events obviously, which can be used as the ideal drugs treatment of UC.
