A Pilot Study Of Pristane And Lipopolysaccharide Induced Lupus Model In ApoE-/- C57BL/6 Mice

ObjectiveIn order to study the pathogenesis of the early-onset ather osclerosis (AS) in patients with systemic lupus erythematosus(SLE),to establish a mixed experimental model in apoE -/-C57BL/6 mice induced by pristane and lipopolysaccharide。Methods36 apoE-/- B6 female mice were randomly divided into pristane control group, pristane model group , LPS control group, LPS Mod elgroup ;While 34 normal B6 female mice were randomly divided in to pristane control group, pristane model gr oup , LPS control group, LPS model group; Single intraperitoneal injection of 0.5ml normal saline for pristiane control group,Single I ntraperitoneal inj ection of 0.5ml pristane for pristane model group;Intraperitoneal inj ection of normal saline twice a week, each 0.2ml for LPS control gro up,intraperitoneal injection of 250mg/L concentration of LPS twice a week,each 0.2ml for LPS model group;Detected the blood serum levels of ANA,Anti-dsDNA antibodies and Anti-ENA antibodies and compared the differences among the groups after 3 weeks;Detected the blood serum levels of ANA,Anti-dsDNA antibodies and Anti-Sm antibodies and compared the differences among the groups after 12 and 19 weeks。ResultsAfter induced for 3 weeks ,ANA of each model group than in the control group, and apoE-/ - the pristane model group than in the normal B6 pristane group increased;Anti-dsDNA antibodies only apoE-/ - the pristane mod el group than in the control group increased;Anti-ENA antibodies apo E-/ - in the LPS model group than the control group and normal B6 L PS model group increased.After induced for 12 and 19 weeks,ANA and Anti-Sm antibodies of pristane model group than in the control group;After 12 weeks,Anti-dsDNA antibodies only normal B6 the pristane mod el group than in the control group increased.ConclusionThe present premitive study established a mixed experimental model in apoE-/-mice induced by pristane and LPS,which are belong to environmental risk factors for SLE and share the tendency to develop AS。The experiment may indicate the strategy & pathway for the further study of early-onset AS in the patients with SLE.
