Objective To explore the influence of resveratrol on TNF-αand IL-1 of dorsal root ganglion neurons in rats with nucleus pulposus allografted into epidural space and study the possible protection mechanism of resveratro1.Methods Forty-eight Sprague–Dawley rats were divided randomly into 4 groups: fat+ normal saline group (FS group, n=12), nucleus pulpus+ normal saline group (NS group, treated with normal saline, ip, n=12), nucleus pulpus+ dexamethasone (ND group, treated with dexamethasone , 25 mg/kg,ip, n=12) and nucleus pulpus+ resveratrol group(NR group, treated with resveratrol, 50 mg/kg,ip, n=12 ). In this study, we harvested the nucleus pulposus from the tails of rats and transplanted it to the peripheral of the same rat's lumbar nerve roots. While the lesions were obtained and studied by conventional histology.7 days and 14 days after surgery, the L4- L5 DRGs were seperated, and the expressions of TNF-αand IL- 1 in DRGs were detected by using immunohistochemistry method. The date was managed by the SPSS 17.0 software.Results 1. Peripheral applying of the nucleus pulpusus to DRGs caused obviously significant endoneurial edema, inflammation reaction and demyelination in the rats. Received resveratrols or dexamethasone by intraperitoneal injection could diminish the inflammation reaction. 2. 7 days after surgery, the expressions of TNF-αand IL-1 in NS group,NR group or ND group was stronger than FE group (P<0.05); but the expressions of TNF-αand IL-1 in ND group or NR group was less than NS group without significant difference compared with FS group (P>0.05) at 14 days after surgery. The expressions of TNF-αand IL-1 in DRGs between ND group and NR group were no significant difference in the two phases. (P<0.05)Conclusion 1. Significant inflammation reaction can be seen in the DRGs after autologous nucleus pulposus implanted. 2. Both resveratrol and dexamethasone could reduce the levels of TNF-αand IL-1 of DRGs in rats with topical application of NP and exert a protective effect against inflammation reaction.
