Ledum palustre L. var. Angustum E. Busch resource is very rich, which mainly concentrates in Daxing'anling area of our country, and also distributes in Xiaoxing'anling and Changbai Mountain. There are many active components in Ledum palustre, and they have important medicinal value and aromatic value. In this paper we established a RP-HPLC method for the determination of aesculetin, fraxetin, hyperoside and quercetin in Ledum palustre, and compared content of the four active components in different area combined with ecological environment factors, then aided analysed component difference with IR. We finally established the optimum extracting technology of the four active components. The results were as follows:1. The RP-HPLC system for the determination of aesculetin, fraxetin, hyperoside and quercetin in Ledum palustre consisted of Inertsil ODS-SP column (5μm,4.6 mm×250 mm), Methanol-0.2% Acetic acid (43:57) as mobile phase. Flow rate was 1.0 mL·min-1. The detector wavelength was set at 350 nm. The sample size was 10μL and the column temperature was 30℃。2. Ledum palustre leaves from different areas had different content of aesculetin, fraxetin, hyperoside and quercetin, and content of the four active components in red flag forest amuer forestry bureau and sajiqi forest genhe forestry bureau were relatively higher. Different parts of Ledum palustre had different content of the four active components, and content of the four active components in Ledum palustre leaves was relatively higher. Active components content affected by climate, terrain, soil and picking time.3. Results of FT-IR spectra and the second derivative IR spectra of Ledum palustre indicated that chemical components of Ledum palustre leaves from different areas and different parts were different. Chemical components contained in flower, leaf and branch of Ledum palustre were much more than those contained in root and seed.4. The optimum extracting technology condition of aesculetin, fraxetin, hyperoside and quercetin from Ledum palustre was:ultrasonically extracted with 9 times amount of 65% ethanol,45 minutes for 2 times.
