Part 1 Literature ResearchDiabetes Mellitus (DM) is a kind of metabolic disease on abnormal insulin secretion, leading to metabolic disorder of carbohydrates, fats, and protein. It is characterized by high blood glucose. In order to prevent and treat T2DM with Traditional Chinese Medicine (TCM), we developed Jiangtangsanhuang Pill on the basis of the classical prescription:Taohe Chengqi Decoction. As for DM as well as its complications, the application of Jiangtangsanhuang Pill has reached remarkable curative effects in the clinic. The aim of this experiment is to find the effective target of Jiangtangsanhuang Pill in depth. Part 2 Experimental ResearchObjective:To observe the influence of antioxidation on Jiangtangsanhuang Pill in liver tissues of diabetic rats. To investigate the curative effects and mechanisms of Jiangtangsanhuang Pill in oxidized mechanism way.Methods:The experimental rats were intraperitoneal injected STZ except for the 10 rats in normal group. The experimental rats were randomly divided into 13 for model group,13 for Jiangtangsanhuang Pill intervention group and 10 for metformin intervention group. Jiangtangsanhuang Pill was intragastric adminstrated as 787.5mg·kg-1·d-1, while metformin was as 157.5mg·kg-1·d-1. Normal and model group were fed up with high cholesterol diet for 8 weeks.①he Fasting Blood Glucose (FBG), cholesterol, triglyceride and insulin levels were detected respectively one week after injecting STZ as well as at the end of the experiment.②At the end of the experiment, the rats were killed and the liver tissues were detected for SOD, MDA, GSH in every group.③At the end of the experiment, the pathological changes of the liver tissue were observed through the light microscope.Results:Compared with normal group, the rats in model group were less active, apathetic, the colour of the fur was grey and yellow and lack of luster hours after modeling, the rats'urine and appetite increased significa in model group, while the weight decreased 48hours after injecting STZ. rats'weight in normal group increased gradually. After 72 hours to one w all the rats'weight came to stable. After one week, the weight in no group and model group had a significant difference (p<0.01); By the en the experiment, the weight in model and western medicine group was lower that in normal group, which had a significant difference (p<0.01); the we in Chinese medicine group was higher than that in western medicine group, had a significant difference. (p<0.01). After one week, the blood glucose cholesterol increased and the Insulin Sensitive Index (ISI) decreased.11 a significant difference comparing with normal group (p<0.05). After 8 we the blood glucose and cholesterol decreased and the ISI increased, whicl" a significant difference comparing with model group (p<0.05).Indicators of antioxidant showed that Jiangtangsanhuang Pill can imp the antioxidant capacity of liver. Its function was the same as metfor SOD index was decreased significantly comparing to normal group (p<0. The MDA in model group was higher than that in normal group, which hac significant difference (p<0.01). Both of the treatment in Chinese medi and western medicine made MDA level decrease, indicating that they he statistical significance, the decrease in western medicine group was r significant (p<0.01). While in Chinese medicine group was p<0.05. GS model group was lower than that in normal group, which had statistical significance (p<0.01), GSH increased in Chinese and western medicine treatment, they both had statistical significance. Chinese medicine groi <0.01) was more superior than which in western medicine (p<0.05).Fatty degeneration of liver cells in model group had significant difference comparing with normal group(p<0.05), so as to Chinese medi group and western medicine group. (p<0.05).Conclusion:㎎iangtangsanhuang Pill can decrease the level of diabetic rats't glucose and cholesterol, improve insulin resistance and diabetic sympt ②In terms of antioxidation, the effect of Jiangtangsanhuang Pill is the same as metformin.③The capacity of improving fatty degeneration in liver cell is more superior than metformin.
