It is a problem in clinic to repair of bone defects caused by varieties factors. Traditional therapy such as autogenous bone graft exist certain flawed. With the development of tissue engineering and molecular bionics, many researches study on the field of biological material. But poor biocompatibility and bone repair capacity restrict the applicant of bone repair material. BMP2 is widely used in clinic as a key growth factor which can not be control released and is easily degradation by enzyme.To solve the problems above, in this study, the design of bioactivity is based on the potential ability to bind and slowly release CS/HA-binding growth factors. To engineer a nano-fibrous scaffold with three-dimensional network structure, in this work, chitosan nano-fibers mimicking the natural ECM fibers were prepared by phase separation.The surface morphology, porosity and whether the BMP2 is binding to the CS/HA of scaffold were characterized by scanning electron microscopy (SEM) and FTIR test respectively. The results showed that CS nano-fibers with BMP2 has a diameter of 50-500nm distributed in each pore and formed three-dimensional network structure. Drug release research indicated the nanofibers were excellent carriers of BMP2 Futhermore the results of C2C12 and material co-culture experiment told us that nanofibers had a good cellular compatibility, while the animal experiments show the biological activity of BMP2 which can promote new bone formation.
