Background and purpose:HIF-1 is a heterodimers consisting of HIF-la subunit and HIF-1βsubunit. The physiological activity of HIF-1 depends on the activity and expression of HIF-la subunit. In most cells, the expression of HIF-1 can be induced by hypoxia, so it can mediate every kinds of hypoxia-related physiological response, including the promotion of oxygen transport, angiogenesis, metabolic adaptation, and cell proliferation/survival and so on. As we all know, PET-CT examination can give the early detection and identification of malignant tumors mainly used with the level of glucose utilization, that is 18F-FDG uptake levels (this level of cellular uptake commonly uses standardized uptake value-SUV to represent). Studies have shown that, HIF-1α,through the following three ways:promoting angiogenesis, increasing glycolysis occurs, and regulating the expression of Glut, plays a key role in the development process of tumor. From which we can learne that HIF-1 could regulate glucose metabolism in tumor, then, whether the level of HIF-1 expression is related with the level of 18F-FDG uptake,literature is still not reported, which is the focu of our experimental study. Through this study, we detecte the HIF-1 expression and suv value of different differentiated gastric cancer; compare the two test results, discuss the relationship between the two.Methods:1. Collect totle 30 cases of high, medium and low differentiated gastric mucous cell carcinoma patients' pathological specimens (including frozen general specimens and the corresponding paraffin) from Nanjing Drum Tower Hospital between January 2009 in January 2011.2. Let the patients do PET-CT examination before operation, and determine its SUV values.3. Use immunohistochemical staining SABC method (by kit instructions steps) to measure the Hif-1αprotein expression and also provide precise localization.4. Through the western-blot methods to determine Hif-1αprotein expressions of the specimens, and compares them with the SUV value to find the relationship.Results:1. The expression and distribution of Hif-la in gastric carcinoma organization The positive expression of Hif-la is in cytoplasm or within the nucleus, shown as tan-yellow particles. In 30 cases of gastric carcinoma organizations,the positive expression of Hif-1αis 76.7 percent(23 cases with positive expression),and there is statistically significant compared with normal tissue (P< 0.01).2. For different differentiation of gastric carcinoma tissues, the positive expression of Hif-1αalso shows statistically significant (P> 0.05).3. In different differentiation of gastric carcinoma organizations, there is positive correlation with the expression of Hif-la and 18F-FDG intake (r=0.875, p<0.01)Conclusion:Combining the examnation of Hif-la with PET-CT could be more accurately response to the biological characteristics of gastric carcinoma, and it will give a guiding significance for diagnosis and tissue-typing of gastric carcinoma.
