To investigate the expression of EP2 receptor and microglia in rats'related encephalic regions of methamphetamine intoxication,combined with earlier results to explore the mechanism of COX-2→PGE2→EP2 receptor and the microglia activation in the methamphetamine intoxication rats.Methods:20 male Sprague-Dawley rats were randomly divided into control group(n=5) and experimental group(dividing it into the first, second and third group after the last injection, n=5).Rats in the experimental group were intraperitoneal injected with methamphetamine hydrochloride (10mg/kg), and those in the control group were injected with saline with the same volume (twice a day at 8:00 and 20:00 respectively for 4 continuous days).Then, observing their behavioral changes.Then,examining the protein expression of EP2 receptor and Ibal of intoxication rats in the related encephalic regions by immunohistochemistry technique and analyzing the test results through image. Results:The SB score and endurance have statistical significance between the control group and experimental group; The EP2 receptor and microglia have the expression in each group. Compares with the control group, at the first day, the expression of EP2 receptor reduced.At the second day, the expression of the expression of EP2 receptor still stay a low level. At the third day, the expression of EP2 receptor become close to the control group. The research has shown obvious statistical significance between control group and ld,2d group and between control group and 3d group has not shown obvious statistical significance;There has shown obvious statistical significance in the 3 experimental group. The expression of microglia is higher than the control group in the 3 experimental group, and the 3d group is higher than the 2d group, the 2d group is higher than the 1d group. The research has shown obvious statistical significance between the control group and experimental group There has shown obvious statistical significance in the 3 experimental group.Conclusion:1.Setting up the model of methamphetamine intoxication; 2.MA can cause the microglia activation;3.The way of COX-2→PGE2→EP2 receptor and the microglia activation have not obvious relevance.
