Objective:(1) To establish a reliable rats model of liver injury with CCL4.(2) To investigate the effects of rat bone marrow stem cells combined a-interferon treatment of rat with chronic liver injury.Methods:Chronic liver injury was induced in 60 rats with a single intraperitoneal injection of 50 % Carbon tetrachloride, twice a week in 6 weeks. The rats with chronic liver injury were randomly divided into control group (C), a-interferon treated group(I), rat bone marrow stem cells treated group(S) and rat bone marrow stem cells combined a-interferon treated group(U). The level of AST, ALT, TBIL, ALP, LN, HA, PIIINP and Collagen Type IV in blood serum were measured on3,6 days post-therapy. At the same time, tissue samples were taken from the liver, fixed in 10% neutral buffered formalin, the tissue were stained with hematoxylin-eosin(H&E) for histopathology observation.Results:(1) There were 54 rats survival after the single intraperitoneal injection of 50% Carbon tetrachloride,twice a week in 6 weeks. The model successful rate was 90%. All rats abdominal circumference were increased. And there were chronic liver injury by observing 6 autopsy rats were selected randomly.(2) After drug treated, there were significant difference in four groups. AST, ALT, TBDL,,LN,HA, PIIINP and Collagen Type IV in the serum decreased with time prolongation. The serum AST, TBIL, ALP and LN level in the group(U) were lower than in the group(S),the level in the group(S) were lower than in the group(I) and in the group(I) were lower than in the group(C) (P<0.01).The serum ALT, HA and PIIINP level in the group(U) were lower than in the group(S), group(I) and group(C) (P<0.01), the level in group(S) were lower than group(I) and group(C),the difference between group(C) and group(I) had no statistical significance. There were significant histological difference between four groups. Hematoxylin-sosin(H&E) for histopathology observation:the chronic injury in group(C) were worse than group(S) and group(I), the mildest injury group was group(U).Conclusions:(1) Establishing rat chronic liver injury with a single intraperitoneal injection of 50% Carbon tetrachloride, twice a week in 6 weeks is stable and reliable.(2) Rat bone marrow stem cells or a-interferon had meaningful results in rat with chronic liver injury. The rat bone marrow stem cells therapy is better than a-interferon.(3) Rat bone marrow stem cells combined a-interferon treatment of rat with chronic liver injury are better than mono therapy.
