Purpose 1.To measurement the content of serum folic acid and vitamin B12 in breast cancer with normal people crowd, and determine their differentiations,To study whether there has relevance between serum folate and vitamin B12 and breast cancer.2. Through testing the gene polymorphism about the Methionine synthase and the Methylene tetrahydrofolate reductase, To study the relationship of breast cancer susceptivity with MTHFR, MS polymorphism, also with drinking and smoking in breast cancer susceptibility that comprehensive effect.Method 1. The detection of Vitamin B12 and serum folate, the experimental group were 95 cases of the breast cancer patients who were initially treatmented. The control group was 90 cases of healthy check-up person. Acquisition research object peripheral venous blood with fasting, applied automatic immune analyzer determines the content of vitamin B12 and serum folate. The contents of Vitamin B12 demonstrate in pg/ml, folic acid content representation of nmol/L2.To analysis the risk of MS and MTHFR gene polymorphism associated with breast cancer. The experimental group were95 cases of breast cancer patients who were initially treatmented, for the control group were healthy check-up person 90 cases. Acquisition research object peripheral venous blood with fasting, After DNA extracted ordinary PCR-RFLP technology was applied to 95 cases of breast cancer patients and 90 cases of normal peoples, and derermine different genotypes. The results were showed with constitutes ratio (%).3.Statistical analysis:SPSS statistical packages were applied to the statistical analysis 115,000. Results 1.Breast cancer vitamin B12 levels higher than that of normal control group (P =0.00), and serum folate below the normal control group (P=0.00).2.This experiment did not find MTHFR 1298 gene polymorphism of the influence of breast cancer alone, but found MTHFR C677T have MTHFR 1298 and joint role, and who carries genotypes of MTHFR-677CT and MTHFR-1298AA,the chance to breast cancer ascend to 1.6 times(95% CI:0.637~2.117,P=0.026),carries MTHFR-677CT with the MTHFR-1298 AC ascending to 1.88 times (95% CI:0.714~4.95, P =0.046), MTHFR-677TT and MTHFR-1298AA carriers was 1.94 times(95% CI: 0.934~4.025,P=0.043).3. There has obvious positive interaction with MTHFR 667 genotype and smoking, alcohol consumption.4. There has no significant statistical correlation between MS A2756G gene polymorphism and breast cancer.Conclution1.There is correlations of the contents of Vitamin B12 and serum folic acid with the breast cancer.2. There has significant correlation with MTHFR gene polymorphism and the breast cancer, also has positive interaction with smoking, alcohol consumption.3. There has no obvious statistical significance between MS2756 gene polymorphism and breast cancer.
