Objective:To study the homing ability of bone marrow mesenchymal stem cells (BMSCs) in ischemic tissue between heterogenic hosts.Methods:1. The isolation and purity appreciation of EGFP-BMSCs:bone marrow mesenchymal cells of EGFP-mice (C57BL/6) inoculated in culture bottle. By way of changing the culrure medium gradually, the cells which didn't stick to the wall may being taken away and get purified EGFP-BMSCs. Then we detect the purity of EGFP-BMSCs by flow cytometry when fifth generation.2. Ischemic injury model:Anesthetize mice, shear lateral limbs and disinfect, to expose the right quadriceps by anterolateral incision, isolate quadriceps and ligate proximal and distal of quadriceps with suture, then stanch and suture incision. Left incision, the quadriceps was not isolated and ligated. The same model was established in other mouse; ischemia model in rats, we separate and ligate the lateral head of quadriceps3. Digest the EGFP-BMSCs from the same generation of one mice, adjust the concentration of EGFP-BMSCs (2×106/ml),0.1ml were injected to subjects through tail vein. On 1st day,4th day,7th day,14th day,28th day,6 subjects were sacrificed, get ischemic muscle tissue and contralateral muscle tissue, extract total protein for Western blotting, and do grayscale scan analysis and statistical analysis.Results:1. Detecte EGFP by Western blotting, EGFP expressed in ischemic side, no in the control side, indicating that after EGFP-BMSCs were transplantated to tested mice, EGFP-BMSCS can home to ischemic tissue.2. After statistical analysis of gray value between the ischemic sides of different hosts, P>0.05, not statistically significant, indicating that the variant EGFP-BMSCs with low immunogenicity. Conclusion:1. BMSCS can home to the ischemic muscle tissue.2. Using BMSCs of EGFP transgenic mouse can be a good tracer of BMSCs'homing.3. There is no difference between species on homing ability of BMSCs, BMSCs with low immunogenicity.4. The homing of BMSCs is an initiative migration.
