For obtaining the stable and efficient influenza vaccine, liposome was used as a new adjuvant to produce the dry power of purified influenza virus split vaccine liposome.Liposome encapsulating with vaccine has the function of adjuvant and carrier.Drug-loaded liposome is an ideal immune adjuvant, after entering the body, majority of them were uptaken by phagocytic cells, and easily focused on the main immunological organs, which is good for delivering the antigen to antigen-presenting cells or other immunological activated cells to activate and enhance the immune response. Pulmonary delivery of liposome vaccine not only has the obvious function of adjuvants to enhance immune response, but also the inductive effect of mucosal, cellular and humoral immunity.Thin film evaporation-lyophilization method was employed in this research to produce dry power of purified influenza virus split vaccine liposome by using phospholipids and cholesterol as raw materials. The particle shape and size distribution were analyzed through microscope observation, the liposome suspension and dry powder's preliminary stability were studied by taking appearance and encapsulation rate as index. In this study, the traditional injection approach of vaccination immunity was changed to pulmonary vaccine delivery to clean Kunming female mince. Using MTT to measure the spleen cell proliferation in vitro; to determine the levels of CD4 and CD8 T-cell and the CD4/CD8 using flow cytometry.The two experiment were employed to measure the effect of the cellular immunity.According to the optimized prescription, the encapsulation rate of influenza vaccine liposome lyophilization powder was 83.4%, the liposome particles were in forms of round or elliptic, at a mean size of 2.14μm, distributed evenly. At the same dose, the mice which took pulmonary delivery of influenza vaccine lyophilized liposome can induce the cellular immunity, and the level of the cellular immunity is higher than that of the influenza vaccine non-liposome immunized through intraperitoneal injection.
