[Objective] Rat bone marrow mesenchymal stem cells (rabbit mesenchymal stem cells, MSC) cultured purified, induced to become bone cells, cardiac cells and islet-like cells, differentiation stages observed cell morphology, detection of specific factors or the expression of markers[Methods] Vitro cultured bone marrow mesenchymal stem cells induced by the literature method of induction, respectively, in the first day of an inverted fluorescent microscope 3,7,14,21 dynamic changes in cell morphology was observed; and by RT-PCR, and enzyme-linked immunosorbent assay Detected at different stages of stem cell transplantation in the induction of specific factors or the expression of markers.[Results] MSC adherent to normal growth, antigen expression is CD34-/CD45-/CD73+/CD 105+. Under inverted fluorescence microscopy cell morphology changed significantly, RT-PCR results showed that GATA-4, Nkx2.5, Sarcomericactin and Connexin-43mRNA myocardial cells to induce the expression of the process of increasing, Isulin 1, Isulin 2, CK-19, Nestin, Glut2 and pdxl mRNA expression in the islet-like cells continued to increase during induction, indicating that the inducing agent able to induce induction into osteoblasts, cardiac cells and islet-like cells.[Conclusion]â‘ FP labeled RMSC established in vitro isolation, cultivation and identification of technology platform.â‘¡established the MSC to the myocardial cells, bone cells and islet-like cell differentiation induced system.â‘¢RT-PCR results showed that MSC successfully into myocardial cells and islet-like cells, and secrete the appropriate cytokines.â‘£enzyme-linked immunosorbent assay showed that MSC-like cells to the islet and osteoblast differentiation, and secretion of insulin and alkaline phosphatase.
