Objective:This study will explore the Fas-1377G>A and the FasL-844C>T polymorphism and primary open-angle glaucoma (Primary Open Angle Glaucoma, POAG) the relevance, and to explore the intraocular pressure, axial length, corneal thickness the impact of POAG.Methods:35 cases of POAG patients and 97 normal eyes in Yunnan were collected. Peripheral venous anticoagulant, using phenol-chloroform extraction of DNA, polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), non-denaturing poly vertical slab polyacrylamide gel electrophoresis, stained with silver nitrate method of Fas-1377, FasL-844 polymorphisms of two genes for genotyping and allele frequencies associated with POAG, as well as age, gender, eye axis, corneal thickness and other factors on the onset of POAG.Results:(1) Fas-1377 3 locus genotype frequencies in the POAG group were17.2% (GG),44.8%(GA),37.9%(AA); in the control group were 13.4%(GG),41.2% (GA),45.4%(AA), the distribution of the difference was not statistically significant (P=0.74). FAS-1377A case group and control group allele frequencies were 39.7%, 34%, FAS-1377G case group and control group allele frequencies were 60.3%,66%, gene frequency in both groups no significant difference (P=0.43,OR=1.27,95% CI =0.697~2.332), cases and controls are in line with the release of Hardy-Weinberg equilibrium test (patients:P=0.73, controls P=0.42). Logistic regression analysis with the dominant model for the Fas, stealth model,2 additive models OR values were 0.66,0.59,0.71,0.5 respectively, suggesting that Fas-1377 gene carriers may reduce the risk of suffering from POAG. (2) FasL-844 genotype frequencies of three loci in the POAG group was 45.5%(CC),31.8%CT.22.7%(TT), the control group were 51.5%(CC),39.2%(CT),9.3%(TT), the distribution of the difference was not statistically significant (P=0.43). FasL-844C case group and control group allele frequencies were 64.3%,71.1%, FASL-844T case group and control group allele frequencies were 35.7%,28.9%, and allele C, T between the two groups There was no significant release (P=0.38, OR=0.73,95% CI=0.361~1.476), cases and controls are in line with the release of Hardy-Weinberg equilibrium test (patients:P=0.209, controls P=0.65). Logistic regression analysis with 4 models, FasL-844 gene carriers may be increased risk of suffering from POAG, OR values were 1.87,2.12,1.66,1.66; P>0.05, not statistically significant. (3) POAG group axial length of the case group and 24.08±1.96 mm, control group 23.16±1.03mm, the difference between the two groups was statistically significant (P=0.017).The corneal thickness between the two groups p=0.91,the difference was not statistically significant.Conclusion:(1) Fas-1377G>A and POAG susceptibility may no significant correlation, FasL-844T> C polymorphism was associated with POAG susceptibility, FasL may be the causative gene in this study population of POAG. (2) POAG in the axial growth of the population increased incidence of myopia.
