Immune privilege in the anterior chamber is a multifaceted phenomenon that may have evolved to protect delicate ocular tissues from immune-mediated damage. So,it has a high success rate. However, immunoloically mediated rejection is the most important cause of transplant failure,especially in high-risk patients.So,it Has very important significance to Establish an appropriate animal model of corneal transplantation for further simulate human corneal transplant rejection, more in-depth basic research.Mechanisms of corneal transplant rejection is a variety of immune cells and immune molecules in the complex process of immune response, Effective prevention and treatment of corneal allograft rejection of corneal transplant rejection is to reduce the rate of the practical problems.NK(Nature Killed cell) is a class of Independent lymphocyte population, Studies have shown that NK cells play the role of recognized allogeneic antigen recognition, and release cytokines to activate the role of different effector cells.Objectives:To establish an appropriate animal model of corneal allograft rejection,Observed the performance and the development process of corneal rejection. Analyze the precautions and skills in the surgery,postoperative complications and Countermeasures, to improve the success rate of corneal transplantation model. To preliminary study the role of natural killer cells in rat corneal allograft rejection.Methods: 1.Construct animal model of corneal transplantation1.1 The experimental group:40 Wistar female rats,40 SD female rats, drill SD rat right cornea with a diameter of 3.5mm Diamond ring as graft for the allogeneic corneal transplantation, drill Wistar rat right cornea with a diameter of 3.0mm Diamond ring as planting bed,then, the corneal graft will be interrupted suture in the corneal bed with 10-0 nylon line.1.2 The control group:40 Wistar female rats, drill Wistar rat right cornea with a diameter of 3.0mm Diamond ring, At the 12:00 mark bit, interrupted suture in situ after rotating 180°with10-0 nylon line.1.3 Observe postoperative:Observe the rejection process of Postoperative corneal graft,and rate reject reaction of corneal transplantation at different time afert surgery.2. pathology research of the rat corneal allograft rejection, Analyze ComplicationsExperimental group and control group were removed four eyes from each group at PO6,PO8,PO12,PO24,PO45.
