(School of Chinese Pharmacy, Beijing University of Chinese Medicine, Beijing 100102)This dissertation included five parts:Part one A review on the research progress of Onocleaceae in chemical constituents and pharmaclogical activities was presented.Part two The study on chemical constituents of Malteuccia struthioptreis(L.)Todaro. Malteuccia struthioptreis(L.)Todaro. was chose as material to study.8 compounds were isolated from sediment and chloroform parts of extract of Malteuccia struthioptreis(L.)Todaro by silica gel. Using the technique of spectrum (1H-NMR,13C-NMR), four compounds were confirmed, They are matteucinol,5,7,3'-trihydroxy-6,8-dimethyl-4'-methoxy-flavanone, desmethoxymatteucino and matteucinol-7-O-glu. 5,7,3'-trihydroxy-6,8- dimethyl-4'-methoxy-flavanone and matteucinol-7-O-glu were firstly found in Malteuccia struthioptreis(L.)Todaro. An fattyalconol compound was also isolated.Part three The quantitative determination of total flavanone in Malteuccia struthioptre is(L.)Todaro.The methodological study on the quantitative determination of total flavonoid in Malteuccia struthioptreis(L.)Todaro. was done by NaNO2-Al(NO3)3-NaOH colorimetry. Using rutin as reference substance and 5%NaNO2-10%Al(NO3)3-5%NaOH as chromogenic agent, checked the absorbency in 494nm to determine the content of total flavonoid. The calibration curve was linear within the range of 9~60μg/ml, the regression equation of total steroids was y=0.0122x+0.0034, r=0.9995. The results of precision, repeatability and stability all met our requirements. The average content of total flavonoid of Malteuccia struthioptreis(L.)Todaro in Xi'an, Changbaishan, e'erduosi and Hebei Anguo were 19.756mg/g,18.514mg/g,17.636 mg/g and 18.834 mg/g respectively.Part four The quantitative determination of matteucinol,5,7,3'-trihydroxy-6,8- dimethyl-4'-methoxy-flavanone and desmethoxymatteucino by HPLC from Malteuccia struthioptreis(L.)Todaro. and other commercial Guanzhong.Establishment of the quanlity controlling methods of matteucinol,5,7,3'-trihydroxy-6,8-dimethyl-4'-methoxy-flavanone and desmethoxymatteucino. Regression equations of matteucinol,5,7,3'-trihydroxy-6,8-dimethyl-4'-methoxy-flavanone and desmethoxymatteucino were respectively:y=2055.5x-17.174(r=0.99995), y=5192.8x-2.2476(r=1.00000), y=3531.4x-0.281(r=1.00000), which showed that the calibration curve was linear within the range of 0.308~1.54μg,0.038~0.19μg and 0.018~0.09μg respectively. The average recovery rate was 98.13%,98.75% and 97.38% respectively. The results of precision, repeatability and stability all met our requirements,showed that the methods were steady and reliable and can be used as the determination methods. The content of Matteucinol and 5,7,3'-trihydroxy-6,8- dimethyl-4'-methoxy-flavanone is 4.2256 mg/g and 0.4538mg/g respectively in Changbaishan. The content of desmethoxymatteucino is 0.1857mg/g in Anguo(Hebei).Part five The fingerprint of Malteuccia struthioptreis(L.)Todaro.Establishment of the fingerprint of Malteuccia struthioptreis(L.)Todaro. the absorbency in 210nm, Relative retention time and relative are of single peak, which makes up 3% or above 3% of the are of all peaks, were determined. In experiments of acuracy and reproducibility, RSD of relative retention time was 0.041%-0.177%a nd 0.026%-2.172% respectively, RSD of relative peak area was 0.594%-3.513% and 0.613%-2.773% respectively. The stability experiment revealed that the sample solution is stable in 21 hours.Data were analyzed by "SOP of Similarity evaluation system for chromatographic fingerprint of TMC(Version 2004A)"software to compare the Malteuccia struthioptreis(L.)Todaro in Xi'an, Changbaishan, e'erduosi and Hebei Anguo were all above 0.95 respectively, the data of Guan-Zhong from 7 different habats were 0.517,0.746,0.580, 0.746,0.796,0.606,0.838 respectively.This dissertation makes a foundation for further research of Malteuccia struthioptreis(L.)Todaro.
