Objective:The study on safety pharmacology of shuangshensu capsule. Observe the effect of Shuangshensu capsule on the central nervous system of the mice. Observe the effects of Shuangshensu capsule on the respiratory system and the cardiovascular system of the normal anesthetized dog.Methods:1, Shuangshensu capsules were chronically administered to three groups mice at three doses of 100,200,400mg/kg(20ml/kg).The control group was administered by the same volume 0.5% CMC-Na.After 50min,the mice were injected with pentobarbital at the dose of 50mg/kg.Observe the effect of Shuangshensu capsule on pentobarbital.2, Shuangshensu capsules were chronically administered to three groups mice at three doses of 100,200,400mg/kg(20ml/kg).The control group was administered by the same volume 0.5% CMC-Na.After 50min,the mice were injected with pentobarbital at the dose of 25mg/kg.Observe the effect of Shuangshensu capsule on pentobarbital.3, Shuangshensu capsules were chronically administered to three groups mice at three doses of 100,200,400mg/kg(20ml/kg).The control group was administered by the same volume 0.5% CMC-Na.After 10,30,60min,observe the effect on the mice general states.After 10,30,60min,observe the effect on the mice indepentent activity number(in 10min).4, Shuangshensu capsules were chronically administered to three groups mice at three doses of 100,200,400mg/kg(20ml/kg).The control group was administered by the same volume 0.5% CMC-Na.After 10 min,observe the effect on the mice functional coordination of movement(the rotating rod falling rates in 1min and 3min).5, Dogs were anesthetized with pentobarbital(30mg/kg).Right femoral artery was intubated,record the SBP and DBP through TP-200T type pressure transducer connected with AP-621 type carrier amplifier.Record standard ECG and HR with AC-601G type ECG amplifier.Record RF and RE with TB-611T type tension transducer.Record simultaneously the indicators with RM-6000 type polygraph after administration 15,30,60,90,120,180min. Shuangshensu capsule were administered to dogs through duodenum at three doses of 20,40,80mg/kg(10ml/kg) one time.The control group were administered with the same volume 5%CMC-Na.Result:1.There was no abvious effect of SSC on the sleep time and containinging sleep time of pentobarbital mice.2.There was no abvious effect of SSC on the sleep number of subthreshold dose mice.3.Compared with the control group,the mice general states and indepentent activity(in 10 min) had no distinct change,in addition,there was neither clear changes betweem before and after SSC4.Compared with the control group, the mice rotating rod falling rates in lmin and 3min had no distinct change.5.Compared before adminitation,the RF and RE and HR and SBP and DBP and ECG of the dogs had no distinct change.Conclusion:According these exeperements,we found that SSC have no abvious effect on the central nervous system of mice,have no abvious effect on the respiratory system and cardiovascular system of dog.
