Objective Study the changes of adrenaline concentration in the blood serum when stress reaction happens,and investigate the correlation between stress reaction and the changes of RPE(retinal pigment epithelium) by observing the ultrastructures of RPE under the stress reaction.All above,it will do the groundwork for the occurrence and development of retinal diseases.Methods Divide 24 healthy male Wistar rats into 4 groups to set up stress-rat models.Every group is binded up to induce stress reaction.Collect the specimen of the rats'blood-serum,and we use ELISA for the quantitative determination of adrenaline in plasma.Collect the data and use the SPSS 13.0 statistics software to analyse the data,then observe the changes of RPE cells by light and electron microscope,finally,arrive at the conclusion.Result(1).The adrenaline concentration of comparison group has not changed obviously during the normal feeding period and it has no statistics sense (P>0.05).(2). Accommodation group has stressed from the first week,and its adrenaline concentration has increased obviously comparing with the beginning of the first week.It has statistics sense(P<0.05).Stop stressing from the third week and the adrenaline concentration declines from the end of the third week but it is higher than the beginning of the first week.(3). Accommodation and stress group has stressed from the first week,and its adrenaline concentration has increased obviously comparing with the beginning of the first week.More longer time,more higher.It has statistics sense(P<0.05).(4).Stress group has stressed from the third week,and its adrenaline concentration has increased obviously comparing with the beginning of the first week.More longer time,more higher.It has statistics sense(P<0.05).(5).Light microscope:RPE has not apparent changes.Electron microscope(JEM-1200EX):Nuclears of PRE cells turn irregular partly.Many cellular organs have patho-changes and mitochondria are tumefied.We can see many pigment granules,and occasionally find endoplasmic reticulums have lightly dilated,which are much more severe along with the increase of adrenaline concentration.Conclusion(1). To keep stressing can make adrenaline concentration increase higher. More longer time,more higher.(2).To stop stressing can make adrenaline concentration decline,but higher than before.(3).Higher adrenaline concentration can induce RPE cells to appear hypoxia when stress reaction happens, and more long more serious.
