DCM is a common long-term complication of diabetes mellitus one of special cardiomyopathy. The morphological development of diabetic cardiomyopathy is stigmatic by myocardial extracellular matrix (ECM) accumulation and myocardial interstitial remodeling. The beginning of diabetic cardiomyopathy is asaconsequence of compositional action of multiple factors, including glycolipids disorders, enhancement of production and release of cytokine factors and growth factors.Tissuetransglutaminase (tTG) belong to glutamines kindred.lt can catalysis the metaglutaminate reaction between glutaminate residue and lysine residue.lt can resistance the degradation of matrix metalloproteinases.tTG was considered as an important factor in fibrosis of tissues and organ.So, tTG may serve as a new ideal therapeutic target for diabetic cardiomyopathy. streptozotocin (STZ) of the expressive condition of tTG on the myocardium of diabetic rats induced and tTG the effect of on ECM accumulate have not been determined.DCM is no effective therapies on till now in the modern medicine. the experimental and the clinical study on preventing and treating on DCM traditional Chinese medicine has shown the promising prospecct. TFGs is composites subgroup traditional Chinese medicine, and it can obviously drop blood fat and blood sugar,and inhibit ECM accumulation of DN.Therefor,?the study of TFGs action on myocardium of diabetic rate and the relative molecular mechanism have not been covered. The aim of this study is to preliminarily decline the the protective domino effects on TFGs of Diabetic Myocardial Lesion of Rats.Object:Study about the effect of tTG on cardiomyopathy of diabetic rats.Preliminarily determine the the protective domino effects of TFGs on Diabetic Myocardial Lesion of Rats and its mechanism.Method:study about include the myocardiac morphological changes, immunohistochemistry and RT-PCR. The parameters evaluation includes:â‘ TFGs effects on the myocardium of diabetic rats structure and the protective domino;â‘¡tTG expression;â‘¢the impact of TFGs on the synthetic mechanism of ECM components.Results:1. By induced STZ Diabetic rats were observed opticthat microscope and electric microscope. Their myocardial structures were obviously altered at 12 weeks. The results showed that the number increased of myocardiac interstitial fibrocyte,?protein substances with positive marked by PAS and the content of fibronectin (FN) and collagenâ… (Colâ… ) all increased (P<0.01, P<0.05). The pathological changes of experimental group treated by TFGs for 12 weeks was alleviated more than obviously diabetic group (P<0.01, P<0.05). Besides TFGs has appreciable effect in regulating blood glucose and lipid concentration.2. At12 weeks,?tTG mRNA and protein expression of myocardium of diabetic rats obviously increased (P<0.01).tTG protein expression of myocardium of diabetic rats were positive correlation with Col I and FN protein expression (r=0.698, P<0.05; r=0.743, P<0.05).After being treated by TFGs for 12 weeks, mRNA expression and tTG protein of experimental group differencely decreased 44.0% and 41.2% than diabetic control group (P<0.01).Conclusions:AT 12 weeks,?the myocardium pathological changes of diabetic rats mainly included that Col I and FN multiplying the reconstitution of myocardial interstitium.tTG is an important profibrotic mediator in pathogenesis of diabetic cardiomyopathy.Through decreasing the expression of tTG,TFGs can make the pathological changes less severe of myocardiac interstitial remodelling.
