| The leukemia is known as"blood cance", it's a common malignant tumor of the hematopoietic system, and it's more often occured in young people, came in the first place of malignant disease of young people, seriously troubled person's lives and threatened human's life. The disease was the result of the patient's hematopoietic system produced a large number of immature white blood cells, there by reducing the number of normal blood cell. In our country, the most occurred disease is acute lymphoblastic leukemia, a disease particular harmful to children.This paper using Polymerase Chain Reaction(PCR) to analysis the blood of normal person's and ALL patients',in target of analysis the differents expression of TALl and transcription factors associated with TALl at the transcription level,then to study the fuction of TALl in hematopoietic cell development.As well as to providing the theoretical basis of treatment to leukemia.The results showed in the process of hematopoietic stem cells develop into mature blood cells,the abnormal expression of TAL1 can cause the leukemia.TALl binding with LSD1/CoREST complex and negatively regulates the expression of complex LSD1/CoREST; in the early stage of hematopoietic stem cells development, GATA2 overexpression to proliferation of blood cells,then later, GATA1 increased expression,which can inhibit GATA2,this is the classic"GATA Switch".In the process, TALl positive regulation GATA2, negative regulation of GATA1.During the late stage of erythroid cells differentiation,GATA2 declined expression, while in the ALL patient, the expression of the TALl raised GATA2 expression, causing incomplete differentiation of erythroid cells, can not developed into mature blood cells, then block the hematopoietic function of bone marrow,causing leukemia.In the process of cell cycle and apoptosis,the transcription factors RunX1,CDK6,hp16 and hp21 are all important Colony-stimulating Factors:RunX1 can both inhibit and promote the expression of blood-related genes, CDK6 can promote cell division, p16 and p21 can anti-apoptotic ,TAL1 all inhibit them at the transcriptional level;GATA2 and CDK6, p21 may be interconnected, the role of this association in the hematopoietic system development may play a role of bridge, which may be through GATA2,TALl can regulate CDK6 and p21. |
