ObjectiveTo demonstrate that if there is protective effects on rats which were given erythropoietin during acute phase of cerebral ischemia.To explore the best frequency of EPO administration and get more effective protection of hippocampal neuron of rats in early period of acute cerebral ischemia.Methods80 male SD rats were randomly divided into 3 groups:normal group, ischemia-reperfusion group and treatment group.In addition to the normal group,the other two groups were made of cerebral ischemia-reperfusion model, meantime the treatment group was divided into three sub-groups.Among the various sub-groups,EPO administration(intraperitoneal injiection) were the same,and doses(3000 IU/kg) were the same.Only the frequency of administration was different from each other.Three-dose group,ischemic instantly to a meeting,6h after ischemia to a meeting as well as 12h after ischemia.Two-dose group,ischemic instantly to a meeting and 6h after ischemia.One-dose group,only ischemic instantly to a meeting.To record and analyze the Behavioral score of rats on 24h after ischemia.The rats in ischemia-reperfusion group and treatment group were used for samples which were used for histological morphology in HE staining and analysis of Bcl-2, Bax expression in hippocamp in immunohistochemical staining.There was also determination of Caspase-3 expression in hippocamp in Western Blot.ResultsNeuroethology evaluation:contrast to the model group,there was a obvio- us improvement about neuroethology in two-dose group and three-dose group, there was statistical significance,(P<0.05);there was no statistical significance in one-dose group(P>0.05);A kind of promoting apoptosis protein,Bax:contrast to the model group,the content of Bax was much lower in the three sub-groups of the treatment group, there was statistical significance,(P<0.05);A kind of inhibiting apoptosis protein,Bcl-2:contrast to the model group, there was a obvious improvement about the content of Bcl-2 in two-dose group, there was statistical significance,(P<0.05);meanwhile the content of Bcl-2 was much lower in one-dose group and three-dose group,there was statistical significance,(P<0.05);Apoptosis terminal protein,Caspase-3:contrast to the model group,the content of Caspase-3 was much lower in two-dose group and three-dose group, there was statistical significance,(P<0.05);contrast to three-dose group,the content of Caspase-3 was much lower in two-dose group,there was statistical significance, (P<0.05);the content of Caspase-3 was much higher in one-dose group than the model group;there was statistical significance,(P<0.05).ConclusionsErythropoietin may protect brain of rats within acute phase of cerebral ischemia.Erythropoietin is able to improve the nerve behavioral score of rats. Erythropoietin is able to protect the hippocampal neurons by the role of inhibiting apoptosis,which may enhance the quantity of Bcl-2, meanwhile reduce the quantity of Bax.Erythropoietin is able to inhibit apoptosis terminal protein-Caspase-3.Two-dose group has obvious advantages.It can be seen,two-dose group is close to the best delivery method.
