ObjectiveThe mouse bone marrow mesenchymal stem cells were induced into insulin producing cells by RPE, the cells were packed by microencapsulation technology, and then they were transplanted into the renal capsule of diabetic mice. In order to investigate the therapeutic effect of microencapsulated IPCs in diabetes mice and the biological effect produced by transplant point or the IPCs.MethodsThe mouse bone marrow mesenchymal stem cells were isolated, passagedand purified. RPE was extracted from rats'pancreases. BM-MSCs were induced by RPE. The expression of insulin in IPCs was detected by immunocytochemistry and dithizone (DTZ)-stainning. The cells were wrapped up by sodium alginate. Streptozotocin was injected into the abdominal cavity to make diabetic mice model. The mice were randomly divided into four groups: A group(empty group); B group (BM-MSCs transplantated); C group; (non-microe- ncapsulated IPCs transplantation);D group (microencapsulated IPCs transplant- ation). They were transplanted into renal capsule. In order to observe the effects of treatment, the level of weight, blood glucose, insulin and C peptide were detected After transplantation, themicrocapsules and kidney were took out. The viability of microcapsules was observed by HE stainning.ResultsCultured bone marrow mesenchymal stem cells were purified.By light microscope,the cells was uniformity, a short spindle or needle-shaped, shading light is strong.Induced by RPE, a week later, the cells became rounded, floated and gathered. The expression of insulin could be seen by Immunofluorescence. Dithizone stainning showed the cells was scarlet. The APA microcapsules were uniform, smooth and toughness. Six-CFDA detection showed that the activity of capsule cell was very strong. After 10 days from transplantation, the mouse's weight of B, C and D goups had increased,especially the D group came up to 26.40±1.39(g), compared with other groups , the result showed they have statistical significance P<0.01). The blood glucose of D group had significant declined to 9.34±1.18(mmol∕L), compared with other groups , the result showed they have statistical significance P<0.01). t he blood glucose of B and C group were had a transient dcreased, while keeped a high level in the A group . The levels of insulin and C-peptide increased in C and D group, D group was more obvious. The numerical result came up to 31.62±1.85(μIU∕ml)and 0.37±0.09(pmol∕ml)compared with other groups , the result showed they have statistical significance P<0.01),. The kidneys and microencapsules were took out ten days after transplantation. The detection of microencapsulated showed that the activity of cells were still high, By HE stainning, the abundant transplanted could be seen between the renal capsule and cortex ,formed a sandwich structure.ConclusionsThe IPCs which had been transplanted into renal capsule could significantly alleviate the symptoms of diabetic mice, reduced blood glucose and improve insulin levels and C peptide ,has a certain effect in the treatment of diabetes, The renal capsule can be a ideal taget for the cell transplantation.
