Objective:To detect the variation of the expression of angiogenesis-related gene VEGFR2, Ephrin-B2, Notch 1 and D114 between colon cancer and normal colon tissue, investigate the correlation of their expression and evaluate the relations of these genes with tumor differentiation, invasion, lymph node metastasis and Dukes'staging age.Methods:The mRNA transcription level of Notch1 and D114 were determined by using fluorescent real-time quantitative PCR in 40 cases of colon cancer tissue and surgical margin tissue. The expression of VEGFR2 and Ephrin-B2 protein were detected by using immunohistochemical staining method in the colon cancer tissues and surgical margin tissues. The relations between these genes and the relationship between their expressions and the pathological data were analyzed.Results:The mRNA transcription levels of Notchl and D114 and the protein expression levels of VEGFR2 and Ephrin-B2 were higher in colon cancer tissues than that of surgical margin tissues. Notchl and D114 transcription levels of mRNA had correlation with tumor differentiation, lymph node metastasis, but have not with age, sex, and pathological type of tumor in colon cancer tissue. The mRNA transcription levels of these two genes had correlation. VEGFR2 and Ephrin-B2 protein expression levels had correlation with tumor differentiation, invasion, lymph node metastasis and Dukes'staging age, but no correlation with age,sex, and pathological type of tumor in colon cancer tissue. The protein expression levels of VEGFR2 and Ephrin-B2 had correlation.Conclusions:The higher expressions of VEGFR2, Ephrin-B2, Notchl and D114 in colon cancer tissues indicate that these genes play important roles in the process of tumor angiogenesis. Controlling tumor angiogenesis by targeting these genes may be a new approach for therapy of colon cancer. The relevance of these genes with tumor differentiation, lymph node metastasis and Dukes'staging prompt that VEGFR2, Ephrin-B2, Notchl and D114 may be indicators to the degree of malignancy of colon cancer and lymph node metastasis and has good pathoclinical diagnostic value.
