Objective:To investigate the effect of NSC transplantation combined with IKVAV-PA on the recovery of motor function of rats with SCI.Methods:A total of 30 SD rats were used to establish SCI models,and then divided randomly into three groups.NSC signed by Brdu were cultured in vitro and examined by immunofluorescence.1% IKVAV-PA was trigger to be gel by the addition of DMEM/F12,which was detected under TEM.At one week following injury,medium,simple NSC and NSC+IKVAV-PA were separately injected into damaged sites. All animals were evaluated on the hind limb behavior with BBB locomotor rating scale,and two rats chosed randomly of every group were performed to observe the recovery of nerve by pathology and immunofluorescence atl,2,4,6,8 weeks post-transplantation.Results:At4,6,8 weeks post-transplantation,motor function of the rat hindlimb was significantly improved in simple NSC group and NSC+IKVAV-PA group(P<0.05),but they are same atl,2 weeks post-transplantation. Pathological sections demonstrated that the area of injury is clear in NSC+IKVAV-PA group.Brdu-postive and NF-200-postive cells,Brdu-postive and GFAP-postive cells were detected in the spinal cord by immunofluorescence.And the number of Brdu-postive and NF-200-postive cells was more than that of simple NSC group(P<0.05).Conclusion:IKVAV-PA promotes NSC differentiation to neurons in vivo,and improves the recovery of motor function of rats after SCI.
