Objective:To obtain the peptide mass fingerprintings of serums of the patients with bone metastasis and no metastasis and to screen and identify the differential serum proteins of patients with bone metastasis after radical mastectomy. To establish diagnostic models for diagnosis of patients with bone metastasis after breast cancer operation. Methods:36 serum samples were collected from patients after modified radical mastectomy from Aug.2009 to Oct.2010 (18 samples from patients with merely bone metastasis and 18 samples from patients without metastasis). All the samples were categorized into infiltrating ductal carcinoma based on our histopathological diagnosis. The differential serum proteins were analyzed by Clinprot system which is consist of Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry (MALDI-TOF-MS) and MB-WCX technique. Characteristic peaks were analyzed and selected by analyses software within Clinprot system. Every group was repeated 2 times. Results:All serum samples were repeated after 5 days, the fingerprintings were similar to the former.4 protein peaks were chosen randomly to compute CV which are less than 30%. The Clinprot system has good repeatability. The peptide mass fingerprintings of serums of the patients with bone metastasis and no metastasis are analyzed by Clinprot system. No differential protein was detected. Conclusions:(1)No differential serum protein exists in patients with bone metastasis and without metastasis. Bone metastasis'bolting by detection of differential serum protein in Peripheral blood may be impossible.
