ObjectObstuctive sleep apnea-hypopnea syndrome is common and may have substantial adverse impact on mental health.OSAHS is a common respiratory sleep disorder characterized by recurrent upper airway collapse during sleep,leading to recurrent asphyxia and sleep fragmentation. Patients with OSA are at increased risk of having motor vehicle crashes, cardiovascular disease, hypertension, and stroke. Timely prevention and treatment can not only improve the quality of life of patients, but also prevention of complications.Several enzymes expressed in vascular tissue contribute to production and efficient degradation of reactive oxygen species.Recent work has shown that ROS of human vascular system mostly derive from NADPH oxidase.ROS produced by NADPH oxidase can interact with proteins,lipids,and DNA,cause oxidative stress and some pathophysiology changes.In activation of the oxidase,the multidomain protein p47phox may play a central role.In this study ,serum concentration of coenzymeⅡand phosphorylation of peripheral blood neutrophil NADPH oxidase subuint p47phox were detected ,in order to explore the role in the occurrence of OSAHS.MethodsThirty male subjects with OSAHS were enrolled in the study,who were underwent diagnostic polysomnography.The standard of diagnosis according to recommended Guidelines for Diagnosing and Treating Sleep related Breathing Disorders in Adults. All patients with OSAHS were divided into three groups: mild(age=39.55±6.32,BMI=27.66±2.27kg/m2) ,moderate(age=37.50±3.76,BMI=26.91±1.25kg/m2) and severe(age=40.95±6.85,BMI=28.16±2.04kg/m2) ,each of them have ten subjcts , ten unrelated subjects were recruited as non-OSAHS control subjects(age=37.65±7.04,BMI=27.23±2.97kg/m2). With the inquiry of case history and related physical examination, all of the subjects had excluded chronic or acute liver, kidney and nerve-muscle disorders, acute and chronic infections, autoimmune diseases, cancer, diabetes, etc. Comparisons of sleep parameters between the groups including apnoea-hypopnoea index (AHI), oxygen desaturation index(ODI),lowest oxygen saturation(LSaO2),serum concentration of NADPH were compared respectively among each group. Western blot experiments were performed to analyze protein levels of peripheral blood neutrophil NADPH oxidase subunit (p47phox).ResultThere was no significant difference with age and BMI among the 4 groups. Compared with the non-OSAHS control group, the levels of NADPH were lower in patients with OSAHS which steadily decreased with the increase in severety. (P <0.05)The phagocyte NADPH oxidase subunit p47phox expression was significantly increased in the OSAHS group than that in the non-OSAHS groups (P <0.05).ConclusionThere is oxidative stress in the patients with OSAHS.The degree of oxidative stress relates to the severity of OSAHS.NADPH oxidase p47phox activation particepates in oxidative stress in the patient with OSAHS.
