Objective:Gastric cancer has a high incidence in various cancers, It has a close relationship with H. pylori. TRAF1 and HDAC6 can reduce various kind of disease and cancer, To investigate whether H. pylori can cause TRAF1 and HDAC6 high espression, find the relationship between H. pylori and gastric cancer.To investigate the mRNA expressions of TRAF1 and HDAC6 in gastric epithelial cells (GES-1) treated with two strains of Helicobacter pylori having different clinical features. And measure the protein expressions of TRAF1 and HDAC6 in clinical cases.Methods:GES-1 cells were cultured with two different strains of H. pylori (NO.14 and NO.2) for 24 hours, and the mRNA expressions of TRAF1 and HDAC6 were quantified by PCR. Immunohistochemistry was used to detect the expressions of TRAF1 and HDAC6 in 113 clinical samples which are infected by H.pylori.Results:1. The mRNA expressions of TRAF1 were 0.000245±7.46E-05,0.000446±0.000123,0.000735±0.000117 respectively in control group, NO.14 H. pylori group and NO.2 H. pylori group, with significant differences (P<0.05). The mRNA expressions of HDAC6 were 0.00126±0.00046,0.00213±0.00046,0.00407±0.0011, respectively in control group, NO.14 H. pylori group and NO.2 H. pylori group, with significant differences (P<0.05). The both expressions were consistent with the gene chip.2. The positive rates of TRAF1 were 34.8%,53.3%,72.7%,88.9% respectively in chronic gastritis samples, intestinal metaplasia samples, dysplasia and gastric cancer samples. Compared with dysplasia, the expression of chronic gastritis had significant differences (P<0.05).And so did chronic gastritis to gastric cancer, intestinal metaplasia to gastric cancer. Chronic gastritis and intestinal metaplasia had no significant difference (P>0.05). And so did intestinal metaplasia to dysplasia, dysplasia to gastric cancer.The positive rates of HDAC6 were 21.7%,30.0%,54.5%,77.8% respectively in chronic gastritis samples, intestinal metaplasia samples, dysplasia and gastric cancer samples. Compared with dysplasia, the expression of chronic gastritis had significant differences (P<0.05).And so did chronic gastritis to gastric cancer, intestinal metaplasia to dysplasia and gastric cancer.Chronic gastritis and intestinal metaplasia had no significant difference (P>0.05). And so did dysplasia to gastric cancer.Conclusion:The expressions of TRAF1 and HDAC6 had differences in chronic gastritis, atrophic gastritis and gastric cancer from gene, transcription and protein. It may be helpful in the prevention of gastric cancer which are infected by H.pylori.
