ObjectiveTo investigate the expression of (3-APP, NSE and S100B after severe traumatic brain injury in rats.MethodsThe animal model of severe traumatic brain injury including DAI(Diffuse axonal injury) and FBC(Focal brain contusion),made in 67 SD rats,then they were randomly divided into control group(n=7), sham-operated group(n=12) and traumatic group at six time points (6h, 12h,24h,7d,3d, and 14d). there were 8 rats in each subgrape Immunohistochemical SP method was used to detect theβ-APP expression level in the damaged brain tissue and enzyme-linked immunosorbent assay method (ELISA) was used to detect NSE, S100 in serum after traumatic injury.ResultsPost-traumatic brain injury, it was noted thatβ-APP expression in traumatic group increased and reached its lowest point at 6h and 12h, and it showed higher than that in the normal and sham-operated group (P< 0.05), after three days reached its peak, then that was reduced significantly after one week, and returned to near-normal levels after 2 weeks (no significant difference, P> 0.05), compared with control group. The value of NSE protein in the Serum in the traumatic group at 6h,12h,24h and 3d after injury were higher than that in the control and sham-operated group significantly (P< 0.05). It showed two peaks, one appeared at 6h after injury, and anather at 3d. Then it reincreased and returned to normal level at two weeks after injury. The 3d value was higher than 24h and 7d value (P< 0.01),14d value and normal group 7d value were not statistically significant (P> 0.05). The value of S-100B protein in the Serum in the traumatic group at 6h,12h,24h and 3d after injury were higher than that in the normal group significantly (P< 0.01). So it was concluded that P-APP could be used as an important indicator of brain damage. The fluctuating changes in the expression of Serum NSE, S100B and their correlation with the injury time could be used for the diagnosis and prediction of brain injury.
