Objective:Laryngeal carcinoma is one of the most common head and neck cancers.How to diagnose this cancer earlier is one of the most important ways to make the patients live longer.And now it`s the most popular to find a useful way to diagnose laryngeal carcinoma. In this study,we detected the prevalence of anti-survivin antibodies and anti-p53 antibodies in sera of all the samples by ELISA, compared the positive difference of each group.And then we discussed the diagnosis value of anti-survivin antibodies and anti-p53 antibodies in sera from patients with laryngeal cancer, analysised the relationship between TAA antibody and clinicopathologic parameters, discussed the effect to predict surgery and recurrence of laryngeal cancer.Methods:Blood samples were collected from patients with laryngeal cancer at our hospital (without any other cancers, no treatment in other hospital ), including 31 samples without treatment, 25 samples at 10 days after surgery,23 samples at 17 days after surgery ,20 healthy blood donors. All patients after surgery were diagnosed histologically negative. After centrifugation (3000r/min,10min), sera were divided into aliquots and stored at-80℃.Anti-survivin antibodies and anti-p53 antibodies were detected by ELISA. The cut-off value designating positive reaction was the mean OD+ 2SD of 20 normal human sera.Statistical analysis of differences in the absorbance of anti-survivin antibodies were done by use of the fisher probabilities in 2×2 table test, the ODs from each group were done by Kruskal-Wallis test, using SPSS 17.0 software. Significance was defined as p﹤0.05.Results:1.The prevalence of anti-survivin antibody in sera from patients before surgery was higher than the sera from patients at 10 days after surgery and the healthy volunteers(P<0.05). We found that the positive samples of T1+T2 were less than T3+T4 a differ significantly, which was different with the past reaserch (P<0.05).2.Anti-p53 antibody levels of all the samples were compared and examined for differences.Our reasurch showed that both the prevalence of Anti-p53 antibody and ODs in sera from patients before surgery were higher than the sera from patients at 10 days after surgery and the healthy volunteers(P<0.05).And there was a significant differenc between T1+T2 and T3+T4(P<0.05). 3.In the panel of 2 TAA antibodies of laryngeal carcinoma,we found the sensitivity was higher than single antibody was detected.Conclusion:1.The sensitivity was low, but the high specificity and positive predictive value, the low prevalence of anti-survivin antibody in sera from patients after surgery,indicated that anti-survivin antibody could be a useful tumor marker for patients with laryngeal carcinoma.2.Anti-p53 antibody may be not only suitable for diagnosis of laryngeal carcinoma,but also a prognostic indicator for patients with laryngeal carcinoma.3.The detection of combination antibodies was more useful than single antibody. The combinated detection of anti-survivin antibody and anti-p53 antibody provided an important way to predict the effect of surgery and recurrence of laryngeal cancer.
