Objective To investigate the effect and the mechanism of D-ribose on myocardial ischemia/reperfusion injury of rat hearts.Methods 24 SD rats were randomly divided into Sham group ,ischemia/reperfusion(I/R) group and D-ribose(Ribose)group (n=8). The hearts in Sham group did not undergo the left anterior descending coronary artery ( LAD) ligation; the hearts in I/R group underwent the LAD ligation for 30 min, then were reperfused for 120 min; the hearts in Ribose group were experienced as same as I/R group except for the rats were administered D-ribose (80 mg/Kg) through tail vein 30 min prior to the LAD ligation. The left ventricular samples of the three groups were prepared for investigation. The expression of CINC-1mRNA was detected by RT-PCR,MPO was detected by ELISA,NF-κB was detected by Western blot,and myocardial tissue sections stained by HE were studied under light microscope.Results The expressions of CINC-1mRNA were 0.10±0.02 in Sham group,0.98±0.05 in I/R group and 0.39±0.03 in Ribose group.The content of CINC-1mRNA in I/R group was higher significantly than that in Sham group(P<0.05), the expression of CINC-1 mRNA in Ribose group was decreased significantly compared with I/R group(P<0.05).The expressions of NF-κB were 0.29±0.02 in Sham group,1.68±0.04 in I/R group and 0.97±0.03 in Ribose group.The expression of NF-κB in I/R group was increased significantly compared with Sham group(P<0.05),the content of NF-κB in Ribose group was lower significantly than that in I/R group(P<0.05).The contents of MPO in myocardial tissue were 1.13±0.03 in Sham group,5.68±0.06 in I/R group and 3.47±0.04 in Ribose group.The expression of MPO in I/R group was increased significantly compared with Sham group(P<0.05),the MPO expression in Ribose group was decreased significantly compared with I/R group(P<0.05).Cardiac HE sections were observed under light microscope:more remarkable histopathologic changes and neutrophil infiltration in I/R group than those in Sham group,less remarkable histopathologic changes and neutrophil infiltration in Ribose group than those in I/R group.Conclusion Exogenous D-ribose might play a protective effect on myocardial ischemia/reperfusion injury of rat hearts. The mechanism could be related to decreasing the expression of NF-κB,CINC-1 mRNA and MPO as well as reducing neutrophil infiltration.
