PART 1 A STUDY OF THE PREVENTING EFFECT OF FUSIDIC SODIUM COMBINING RIFAMPICIN AGAINST FORMATION OF STAPHYLOCOCCUS EPIDERMIDIS BIOFILM IN VITROObjective :Established the model of Staphylococcus epidermidis biofilm in vitro,To investigate the preventing effect of fusidic sodium combining rifampicin against formation of Staphylococcus epidermidis biofilm in vitro.Methods: Staphylococcus epidermidis biofilms were prepared with Coverslip incubated with Staphylococcus epidermidis for 24 hours and observed with an optical microscope.Tube fold dilution was used to detect the minimum inhibitory concentration (MIC) of fusidic sodium and rifampicin on Staphylococcus epidermidis.Checkerboard method was used to detect the minimum inhibitory concentration (MIC) of fusidic sodium combining rifampicin on Staphylococcus epidermidis and Calculated Fractional inhibitory concentration (FIC).The effect of fusidic sodium combining rifampicin or alone on Staphylococcus epidermidis was evaluated under the minimum drug concentration.The visible light semiquantitative method was used to measure biofilm value (OD550), To investigate the preventing effect of fusidic sodium combining rifampicin or alone against formation of Staphylococcus epidermidis biofilm in vitro.Results:There are some Staphylococcus epidermidis Adhesion and biofilm formation on the Coverslip.rifampicin or fusidic sodium Significantly inhibited the adherence of Staphylococcus epidermidis and the formation of biofilm compared with the control.Their combination is better than the two alone (P <0.05)Conclusion : rifampicin combined fusidic sodium Significantly inhibited the adherence of Staphylococcus epidermidis and the formation of biofilm compared with the control. PART 2 A STUDY OF EFFECT OF FUSIDIC SODIUM COMBINING RIFAMPICIN IN STAPHYLOCOCCUS EPIDERMIDIS BIOFILM IN VITROObjective :Established the model of Staphylococcus epidermidis biofilm in vitro,To investigate the effect of fusidic sodium combining rifampicin in eradicating Staphylococcus epidermidis biofilm in vitro.Methods:Staphylococcus epidermidis biofilms were prepared with Coverslip incubated with Staphylococcus epidermidis for 24 hours, after the early and maturity biofilm was treated by fusidic sodium, rifampicin respectively, and by the combination, the number of viable bacteria were measured by serial dilution after 4 hours,8 hours,24 hours, the early and maturity biofilm was treated by fusidic sodium, rifampicin respectively, and by the combination were staininged by SYTO9/PI , Observed the number of Dead and live bacteria by Confocal laser scanning microscopy(CLSM). To investigate the effect of fusidic sodium combining rifampicin in eradicating Staphylococcus epidermidis biofilm in vitro.Results:rifampicin or fusidic sodium Significantly reduced the number of bacteria and added to the proportion of dead bacteria in the biofilm. Their combination is better than the two alone (P <0.05).Conclusion : Fusidic sodium combined rifampicin significantly eradicated early and maturity Staphylococcus epidermidis biofilms.
