Objective:To study the treatment of high intensity focused ultrasound( HIFU ) in synaptophysin(SYP) and N-methyl-D-aspartate receptors-2A(NMDAR2A) of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods:One hundred and ninety-two 7-day-old SD rats were randomly divided into four groups: 1 sham group(n=48,A),2 HIBD model group(n=48,B),3 acupuncture group(n=48,C),4 high intensity focused ultrasound group (n=48,D),every group respectively divided into 1w,2w,3w subgroups (n=8).The expressions of synaptophysin(SYP) and N-methyl- D-aspartate receptors-2A(NMDAR2A) in Hippocampus were examined with immuno- histochemical staining and RT-PCR.Results:1. Sham operation group:the structure of brain tissue is clear,the edges of cells were normal.The nuclei were present,and the nucleolus was obvious; HIBD group:the structure of brain tissue is not clear,and the edges of cells were disorder,neuronal necrosis or pyknosis in hippocampal region was apparent with the proliferation of glial cells obviously,Nissl bodies disappear;EA group and the ultrasound group were also have some neuronal degeneration and necrosis,but there are still have the normal structural ,without significant proliferation of gliocytes.2.In high intensity focused ultrasound group and acupuncture group, SYP expression levels were significantly higher than those in sham group and HIBD model group(P<0.05),while the NMDAR2A levels were obviously lower, which had distinct differences compared with the HIBD model group. The expressions of SYP and NMDAR2A in high intensity focused ultrasound group had no difference compared with acupuncture group.Conclusion:High-intensity focused ultrasound therapy can enhance the expression of SYP level and reduce the expression level of NMDAR2A,which has neuroprotective function.Its possible mechanism is that when HIFU is focused on the point,it can play a similar role like acupuncture which can increase the synaptic of HIBD.
