The Association Study Between Brain-derived Neurotrophic Factor Genetic Polymorphism And Parkinson's Disease And Major Depressive Disorder

ObjectiveExperimental and clinical data suggest that genetic variations in Brain-derived neurotrophic factor (BDNF) gene may increase the risk for Parkinson's Disease (PD) and Major Depressive Disorder (MD-D). The present study aimed at exploring the relationship between BDNF and PD and MD-D.Method266 PD patients,83 MD-D patients and 400 controls were recruited for this study, assessed using a battery of neuropsychological tests. According to criteria of the diagnosis of MD-D from Diagnostic and Statistical Manual of Mental (DSM-IV-TR) and the scores of 24 item-Hamilton depression rating scale (HAMD-24), apart the PD group into PD depression group (PD-D group) and PD non-depression group (PD-nD group) and genotyped for 11757C/G, 712A/G, 196A/G, and 270C/T in BDNF gene for all subjects.Results(1) BDNF 712A/G AG genotype was associated with 2.50-fold time risk of PD [odds ratio (OR) = 2.50, 95% Cofidence Interval (CI) = 1.58-3.96].(2) By combining genotypes AG/AA genotypes with 712 GG genotype as reference (OR = 1) in stratification analysis, AG/AA genotypes were associated with PD (2.94, 1.88–4.61).(3) Accordingly, the A allele was significantly overrepresented in PD compared with the G allele (3.16, 2.08–4.81). (4) This distribution of BDNF 712A/G AG/AA genotypes in females and males of PD patients were similar, thus BDNF 712A/G AG/AA genotypes were risk factors for both males and females in PD patients.(5) This distribution of BDNF 712A/G AG/AA genotypes in PD-D patients (3.37, 1.92-5.90) and PD-nD patients (3.54, 1.94-6.47) were similar.(6) BDNF 712A/G AG/AA genotypes only correlated with PD, but not depression symptoms in PD patients. BDNF 712A/G was not the common pathology of PD and MD-D.ConclusionOur results suggested a novel association between BDNF 712A/G AG/AA genotypes and PD in a Chinese Han population. BDNF 11757C/G,712A/G,196A/G,270C/T were not significantly associated with MD-D. BDNF 11757C/G,712A/G,196A/G,270C/T were not the common pathology of MD-D and PD, and was not associated with PD related depressive symptoms.
