Objective: This study was aimed to investigate the expression level of CCAAT/enhancer binding protein alpha gene in patients with myelodysplasyic syndromes (MDS), and to explore the correlation between C/EBPαexpression and MDS.Method: Clinical and laboratory data of 33 cases of adult patients with MDS were retrospectively analyzed, a real time quantitative reverse transcriptase polymerase chain reaction(RQ-PCR)method was used to detect the expression level of C/EBPαmRNA in bone marrow mononuclear cells (BMMNCs) and to investigate the change of expression levels in the pathogenesis of MDS. Combined with The incidence of acute leukemia of C/EBPαhigh expression and low expression groups, we explored the relationship between C/EBPαexpression and disease progression.Results: The expression level of C/EBPαwas significantly lower in low-risk and high-risk MDS than those of normal controls(P<0.01, P<0.001, respectively), moreover, high-risk MDS showed lower C/EBPαexpression than those of low-risk MDS(P<0.05). The acute leukemia(AL) transformation rate was significantly higher in C/EBPαlow expression group than those of high exptession group(χ2=9.17, P<0.05). C/EBPαexpression level in MDS was not correlated with sex, age and peripheral blood cell, while the ratio of blast cell in bone marrow was significantly higher in the C/EBPαlow expression group than in the high exptession group(P<0.01).Conclusions: 1.Reduced C/EBPαexpression level have closely associated with the pathogenesis of MDS, it may be an important molecular biological marker of MDS; 2. the ratio of blast cell in bone marrow was significantly higher in the C/EBPαlow expression group than in the high exptession group; 3.The acute leukemia(AL) transformation rate was significantly higher in C/EBPαlow expression group than those of high exptession group, the degree of down-regulated C/EBPαhas closely related to the progression of MDS.
