Therapeutical Effect Of Hesperidin On Collagen-induced Arthritis In Rats And Its Mechanisms

Objective To study the therapeutic effect of hesperidin (HDN) on collagen-induced arthritis( CIA ) in rats and its mechanisms.Methods CIA was induced with Collagen II and Freund's complete adjuvant. The effect of HDN on CIA was evaluated by body weight, polyarthritis index,hind paw swelling and histopathologica changes. Interleukin-17( IL-17) in serum was examined by enzyme linked immunosorbent assay ( ELISA).Splenic lymphocyte proliferation response induced by concanavalin A (conA) or lipopolysaccharide ( LPS ) was examined with MTT assay. The lever of Phagocytosis of PMΦwas measured by neutral red method.Interleukin-1( IL-1) and tumor necrosis factor-α( TNF-α) in PMΦof CIA rats were measured by ELISA. And the mRNA expression of TNF-αwas detected by RT-PCR.Results1. HDN(80,160,320 mg/kg) singnificantly inhibited the paw swelling and increased body weight of CIA rats. The polyarthritis index and pathological damage of knee joint was reversed by HDN(80,160,320 mg/kg).The index of spleen and thymus were increased by HDN(160,320 mg/kg).The level of IL-17 in serum was decresed by HDN(160,320 mg/kg).2. The enhanced lymphocyte proliferation in CIA rats was reversed by treatment with HDN. HDN ( 10-6 ,10-5,10-4 mol/L)singnificantly reduced the phagocytosis of neutral. HDN (10-4 mol/L) markedly reduced IL-1 .Meanwhile, HDN ( 10-6 ,10-5,10-4 mol/L)singnificantly reduced TNF-αlevel and inhibited the expression of TNF-αmRNA in PMΦ.Conclusion HDN has marked therapeutic effection on CIA rats,which is related to reducing the phagocytosis and inhibiting the secretion of pro-inflammatory cytokines.
