Objective To investigate the relations between the methylation of RAR-βgene, genetic and to evaluate the clinical significance, and to explore the mechanism of lung cancer and offer an effective means for the earlier diagnosis of the malignancy.Methods The methylation of RAR-βgene in DNA from 80 non-small cell lung cancer tissues and corresponding nomalignant tissues were tested with methylation-specific PCR(MSP).Results It extracted genomic DNA from lung cancer tissues and adjacent tissues of lung cancer patients as a template for MSP amplification PCR results showed that 80 cases samples of lung cancer tissues and corresponding adjacent tissues detected at the same time, and 46 cases had methylated positive fragments in lung cancer tissues, 14 cases had methylated positive fragments in adjacent tissues. Their methylation rates were 57.5% and 17.5%. The difference of the methylation rate between the groups was statistically significant(P<0.01). The difference of RAR-βgene methylation rates between clinical staging o~I and II~IV of lung cancer was no significant(P>0.05), and the same to the comparison of central lung cancer and peripheral lung cancer; Different pathological type of lung cancer tissues RAR-βgene methylation test results showed that RAR-βgene methylation detection rates in squamous cell carcinoma were higher than the detection rates in adenocarcinoma and the others, the difference was statistically significant(P<0.05).Conclusion MSP is a simple, sensitive and specific method of detecting the methylation status of CpG-rich region. There was RAR-βgene promoter CpG island methylation in lung cancer tissues, promoter CpG island methylation is closely with the development of lung cancer. The detection of RAR-βgene methylation is contribute to the diagnosis of lung cancer; RAR-βgene CpG island methylation rates of lung cancer isn't related to age, sex, clinnical stage and clinical classification, but may be related to pathological type and smoking.
