Objective: Vascular endothelial growth factor (VEGF)which were named Vascular permeability factor can induces angiogenesis, vascular permeability.it is one of the strongest factor in inducing angiogenesis. it can produced from keratinocyte. VEGF include VEGF121,VEGF145,VEGF148,VEGF165,VEGF165b,VEGF183,VEGF189,VEGF206 . in adult normal skin,VEGF can maintains microvessel density and transporting nutrient substance by blood vessel ,in previous studies, VEGF detected in the lesions of atopic dermatitis is higher than the normal skin . in epidermis tissues, VEGF 121 and VEGF 165 are mainly detected. VEGF may is relative with the serious degree of pathogenetic condition.but mechanism of action is not clear.To study which signaling pathways activated by differential isoforms of VEGF.the differential isoforms of VEGF can induce the different biological effect.The further investigation may explain the relation of VEGF with the atopic dermatitis.It may provide theoretical evidence for whether VEGF is a new target for preventing and treating the atopic dermatitis.It may establish fundament for highlighting mechanism and progression of the psoriasis and atopic dermatitis.Method:1 Human umbilical vein endothelial cells(HUVEC) were cultured in vitro.2 The vein endothelial cells were activated by VEGF 121 or VEGF 165. we retrieved the vein endothelial cells for different time.3 We examined FAK and phosphorylated FAK in the FAK signaling pathway by western Blot . the changes of FAK phosphorylation was analysised.4 We used the same method to examine Akt and phosphorylated Akt in the Akt signaling pathway of vein endothelial cells.the changes of Akt phosphorylationw was analysised. 5 These date were Analysised by statistical software of SPSS13.0Result:1 The expression of phosphorylated FAK in the human umbilical vein endothelial cells after VEGF 121 or VEGF 165 treatments.1.1 The analysis of phosphorylated FAK in the human umbilical vein endothelial cells after VEGF 121 treatmentsThe relative amount of phosphorylated FAK was found in a time-dependence manner from 0min up to 30min when VEGF 121 treated the human umbilical vein endothelial cells , The relative amount of phosphorylated proteins is represented as percentage of total FAK ,it marked increase from 0min up to 30min . 5min0.893±0.048, it was largest in 30 min 1.192±0.151, 60 min 0.966±0.069 The expression of phosphorylated FAK was different in different time(P<0.05).1.2 The analysis of phosphorylated FAK in the human umbilical vein endothelial cells after VEGF 165 treatmentsThe relative amount of phosphorylated FAK was found in a time-dependence manner from 0min up to 10min when VEGF 165 treated the human umbilical vein endothelial cells,The relative amount of phosphorylated FAK is represented as percentage of total FAK .it marked increase from 0min up to 10min . 5min0.469±0.042, it was largest in 10 min 1.015±0.120, The expression of phosphorylated FAK was was different in different time) (P<0.05).1.3 Phosphorylated FAK were compared in the human umbilical vein endothelial cells after VEGF 165 or VEGF 121 treatmentsThe changed tendency of the relative amount of phosphorylated FAK in human umbilical vein endothelial cells which were stimulated by VEGF 121 or VEGF 165 was different (P<0.05). VEGF121 more significantly increased the level of phosphorylated FAK thanVEGF 165,moreover phosphorylated FAK rapidly increased when the human umbilical vein endothelial was treated by VEGF121 than VEGF 165.2 The expression of phosphorylated Akt in the human umbilical vein endothelial cells after VEGF 121 or VEGF 165 treatments.2.1 The analysis of phosphorylated Akt in the human umbilical vein endothelial cells after VEGF 121 treatmentsThe relative amount of phosphorylated Akt was different when the human umbilical was treated by VEGF 121 for different time , The relative amount of phosphorylated proteins is represented as percentage of total FAK , (P<0.05).2.2 The analysis of phosphorylated Akt in the human umbilical vein endothelial cells after VEGF 165 treatmentsThe relative amount of phosphorylated Akt was different when the human umbilical was treated by VEGF 165 for different time , The relative amount of phosphorylated Akt is represented as percentage of total FAK , (P<0.05)2.3 phosphorylated Akt was compared in the human umbilical vein endothelial cells after VEGF 165 or VEGF 121 treatmentsThe changed tendency of the relative amount of phosphorylated FAK in human umbilical vein endothelial cells which were stimulated by VEGF 121 or VEGF 165 was not different (P>0.05). the outputs fited Mauchly of Sphericity(P>0.05).Conclusion:1 VEGF 121 and VEGF165 can activates the FAK pathway, VEGF 121 activates more intensely than VEGF 165, VEGF 121 is predominante isoforms.2 While VEGF 121 and VEGF165 can activates the Akt pathway ,but the changed tendency of the relative amount of phosphorylated FAK in human umbilical vein endothelial cells which were stimulated by VEGF 121 or VEGF 165 was not different.3 With previous studies,It indicates that VEGF 121 which is predominante isoforms induces vascular permeability by FAK and Src signaling pathway...
