Puerariae lobatae caulis and Puerariae lobatae folium is respectively the dried stem and leaf of Pueraria lobata (Willd.) Ohwi. in Leguosae. Puerariae lobatae caulis can clear heat-toxicity, remove edema for relieving pain. Puerariae lobatae folium can arres bleeding.It is researched textually by materia medica that both Puerariae lobatae caulis and Puerariae lobatae folium has a long history for medicinal usage. Puerariae lobatae folium was firstly recorded in Annotation of Materia Medica, Puerariae lobatae caulis was firstly recorded in New Materia Medica which are mostly recorded as the addition of Puerariae lobatae radix in ancient materia medica. Contemporary study about them mostly focus on the chemical component in them while the study on their quality and their pharmacology or potency is rarely reported.Pueraria lobata widely spread in most areas of China. It mainly comes from Henan, Hunan, Zhejiang and Sichuan province. Both the wild and homegrown resource of Pueraria lobata are aboundant.Nowadays, the development and utilization of Pueraria lobata mainly concentrate on its root and flower while its stem and leaf are usually discarded, which leads to the waste of this resource. Pueraria lobata is a deep rooting plant whose developed vertical and horizontal root system are criss-cross in the soil. Therefore, it can strongly fasten soil to prevent the surface soil loss caused by rain wash. However, due to ever-increasing market demand for Puerariae lobatae radix, extensive excavation not only has severely destroyed the wild resource, but has caused the damage of ecological environment. So the study and development on the discarded part of Pueraria lobata (its stem and leaf) is meaningful and will bring economical and social benefit.This project are mainly about Puerariae lobatae caulis'and Puerariae lobatae folium'morphology, chemical component and pharmacodynamics which are as follows:The results of study on Puerariae lobatae caulis'and Puerariae lobatae folium' morphology identification showed that Puerariae lobatae caulis'appearance characteristics are the color of its stems, the shape and arrangement of lenticel, fracture surface's characteristics and the odor; Puerariae lobatae caulis'microstructure characters are the type of cortex cell and the type of inclusion in it, the shape and arrangement pattern of pericycle fiber, the distribution of secretory cell containing roux substances, cambium, the arrangement of ducts and its diameter; Puerariae lobatae caulis'powder identification characteristics are fiber and lens fibers, the type of ducts, prismatic crystals, sclereid, roux pigment bulk. Puerariae lobatae folium' microstructure characters are the type arrangements of nonglandular hair, stoma and vascular bundle of main vein, the distribution of secretory cell. Puerariae lobatae folium'powder identification characteristics are nonglandular hair, type of stoma and prismatic crystals. These characteristics are very evident and are meaningful forPuerariae lobatae caulis'and Puerariae lobatae folium'identification, which provide scientific references for their identification.The TLC identification method for different parts of Pueraria lobata have been established in which Puerarin, daidzin and daidzein are used for comparison. The results showed that the spots of radix and stem are similar in numbers and size, which is correspondent to the report, while the spots of leaf are fewer. The shade of color hint the amount of Puerarin, daidzin and daidzein.Systematical routine inspectations of the Puerariae lobatae caulis and Puerariae lobatae foliumfrom Sichuan have been performed for the first time. The deteration method for extraction content have been established. The results hint that the ammount of extraction content is closely related to the thickness of stem; the content of ash and acid-insoluble ash are different with the circumstance changing. This provide scientific references for the quality evaluation and further development and utilization of Puerariae lobatae caulis and Puerariae lobatae foliumfrom Sichuan.Through the comparison of the content of extraction, total flavone, Pueraria, daidzin and daidzein in Puerariae lobatae caulis and Puerariae lobatae folium harvested from different period, the best picked period forPuerariae lobatae caulis and Puerariae lobatae folium is established. Puerariae lobatae caulis is harvested in the period from late October to midmonth of November when the content of index component is relatively higher, which is correspond with the reports. Puerariae lobatae folium'best harvest period is from midmonth to late of July when the content of index component is relatively higher.Comparing to Puerariae lobatae radix,to exploer initially the effects of Pueraiae lobatae caulis on anti-inflammation and anti-hypoxia activity,and to assess the security of Pueraiae lobatae caulis,which can offer scientific evidences for further study of Puerariae lobatae caulis.the maximally tolerated dose of Pueraiae lobatae caulis is 200g·kg-1,which is 400 times to that andults'. compared with normal saline group,the swelling was inhibited remarkably,and depended on does. compared with the group of Pueraiae lobatae radix,there was no significant difference.both Pueraiae lobatae caulis and Pueraiae lobatae radix could prolong obviously the living time of mice under the normal pressure and hypoxia,and the effcet of pueraiae lobatae caulis was stronger than radix.so,the toxicity of Pueraiae lobatae caulis is small, the effect of anti-hypoxiad and anti-inflammad similar with Pueraiae lobatae caulis.The MICs of three kinds of Pueraria lobata caulis extracts against twenty MRSAs which are detached from pathogens, were performed by agar dilution method.The MICs range of different extracts against MRS A are respectively,40% alcohol extract was from 10.4mg/ml to 83.5mg/ml, in which 20.8mg/ml was in the majority; water extract was from 20.8mg/ml to 83.5mg/ml, in which 41.8mg/ml was in the majority; and 95% alcohol extract was from 41.8mg/ml to 83.5mg/ml, in which 83.5mg/ml was in the majority, three kinds of Pueraria lobata caulis extracts show anti-MRSA activity,and 40% alcohol extract show strongest anti-MRSA activity.
