Objective:1. To analyze the clinical significance of the expression of B7-H1 in clear cell renal cell carcinoma;2. To study the effects of IL-2,IFN-αand IFN-γon B7-H1 expression in clear cell renal cell carcinoma 786-0 cells.Methods:1.The B7-H1 mRNA and B7-H1 protein expression in renal cell carcinoma and the nearby renal tissues was determined using RT-PCR and immunohistochemistry assay in 80 patients,the relationship between the expression level of B7-H1 and clinicopathological characteristics was analyzed;2.786-0 Cells were cultured in RPMI-1640 supplemented with 10% fetal calf serum,The B7-H1 mRNA was detected by RT-PCR and the protein of B7-H1 was detected by cytoimmunochemistry staining,cytoimmunofluorescence staining and flow cytometric analysis. The expression levels of B7-H1 mRNA and the protein of B7-H1 in 786-0 cells were compared.Results:1.The B7-H1 mRNA levels were significantly increased in renal cell carcinoma tissues compared with the nearby renal tissues(P<0.01); The positive rate of B7-H1 in renal cell carcinoma tissues was (33/80)41.25%, the expression level of B7-H1 is relate to clinicopathological characteristics ;2.RT-PCR test showed that each of IL-2 (0.75±0.06),IFN-α(0.68±0.05) and IFN-γgroup (0.95±0.08) of B7-H1 mRNA expression was higher than unstimμlated group(0.30±0.03)(P﹤0.05); The protein of B7-H1 was found to be distributed in the cell membrane through techniques of cytoimmunochemistry staining and cytoimmunochemistry staining, the expression of B7-H1 was upregulated after IL-2,IFN-αand IFN-γstimulation; FCS assay showed that each of IL-2 (65.70±3.26)%,IFN-α(56.52±1.75)%, and IFN-γgroup (84.05±3.52)% of B7-H1 protein expression was higher than unstimμlated group(20.49±1.03)%(P﹤0.01).Conclusion:1. B7-H1 may be used as a new marker to diagnose renal cell carcinoma on early stage and to predict prognosis;2. IL-2,IFN-αand IFN-γcan upregulate the expression of B7-H1 at the levels of mRNA and protein.
