BackgroundHomeodomain interacting protein kinase 2 (The homeodomain-interacting protein kinase -2 HIPK2) can enhance the activity of protein homologous transcription , phosphorylate different downstream targets to activate the apoptotic process, and is also involved in tissue and organ formation of tumors and development. However, at present its expression in tumor tissue is poorly known.ObjectiveTo investigate the homeodomain interacting protein kinase 2 (HIPK2) gene expression level and its clinical significance in non-small cell lung cancer patients . Define wether the levels of HIPK2 impact on the prognosis of patients or hipk2 gene affects the proliferation and migration of lung cancer cells, besides cell cycle distribution.MethodsUsing real-timePCR ,we detected of the levels of hipk2 in the cancer tissues and adjacent tissues of non-small cell lung cancer patients and analysed the relationships between the levels of hipk2 and the clinical and epidemiological characteristics of patients. GFP tagged lentiviral-hipk2-RNAi vector was transfected into H1299,95C and A549 cell lines, aiming to investigate the effection of hipk2 inhibition on lung cancer cell lines.ResultsExpression levels of hipk2 in cancer tissues were lower than that in adjacent tissues; The levels of hipk2 was significant different between smoking group and the non-smoking group (P = 0.024),while between female group and male group(P = 0.021), less than or equal to 50-year-old cancer patients and over 50 years of age (P = 0.046); Patients with low levels of hipk2 may encounter poor prognosis. hipk2 gene silencing enhanced proliferation and migration capacity of the NSCLC cell lines and increased cells G2 / M phase rate by delaying cell cycle progression .ConclusionThe levels of hipk2 in Non-small cell lung cancer tissue and adjacent tissues wree different. Expression levels of hipk2 correlate with the gender, age and smoking and was an important prognostic factor. hipk2 affects proliferation and migration of tumor cells and the cycle. There's evidence that hipk2 is likely to be a tumor suppressor , and can be a useful target for clinical treatment.
