Morphological Effects Of Target Intervention Of The Effective Sites Of Diaomo Zhibengye (Endometrium-removing And Bleeding-stopping Solution) On Endometrial Hyperplasia In Rats

Purpose:The present paper is to observe the effects of Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) on weight, uterine wet weight, index of uterus, endometrial thickness and the microstructures under light and electron microscopes of rat models with endometrial hyperplasia. It is to discuss the targeted interventional effects of Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) on endometrial hyperplasia from the morphological perspective and determine the dose-response relationship, thus to provide theoretical evidence for clinical medication in the future.Methods: 40 healthy female adult SD rats were randomly divided into four groups, which were respectively blank group (Group A), model group (Group B), model + large dose group (Group C) and model + small dose group (Group D). Rats in group A were done opening and closing abdominal wall operation. Rats in group B, C and D had bilateral ovaries been removed. From the third day on, 0.15mL/100g high-pressure sterilized peanut oil was intramuscularly injected into the experimental rat once every other day and 14 times altogether. Then, the rat was killed when the vaginal smear has indicated the estrous cycle. Three days after surgery, rats in group B, C and D were intramuscularly injected 15μg/100g oestradiol benzoate diluted by high-pressure sterilized peanut oil once every other day and 14 times altogether. Thus, rat models with endometrial hyperplasia were built. Later, normal saline solution was injected into each uterine cavity in group B. In group C, 0.3 ml Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) (10%) was injected into each uterine cavity of the rat. And in group D, 0.3 ml Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) (20%) was injected into each uterine cavity of the rat. Three days later, the experimental rat was killed and the uterus was taken and the wet weight was weighed. Then, tissue section was done and the endometrial morphology was observed under light and transmission electron microscopes (TEM).Findings: 1. Rats in group A and group B, C and D have significant differences in terminal weight, uterine wet weight and index of uterus (P﹤0.01). Group B, C and D have no big difference (P﹥0.05). The endometrial thicknesses of rats in group A are far different from group B (much higher) (P﹤0.01). Group A and B have significant difference in endometrial thickness from group C and D (P﹤0.01). There is no big difference between group C and group D (P﹥0.05). 2. Findings of observations under light microscope: Group A: few glands with scattered distribution, small, straight and narrow glandular cavity, cube-shaped glandular cells. Group B: more glands, hyperplasia of glandular epithelium cells with zigzag shape, occurrence of pseudo-stratified phenomenon, increased glandular curvature. Group C: most mucosal epithelia were necrotic and scaled; glands scattered; inflammatory cells infiltrated the muscular layer and serosa. Group D: all mucosal epithelia were necrotic and scaled; glands disappeared; muscular layer bled and swelled; inflammatory cells infiltrated significantly. 3. Ultra microstructure of endometrial tissue under TEM: Group A: Micro-villi on the surfaces of secretory cells were clear. Mitochondria, rough surfaced endoplasmic reticula, free ribosomes, Golgi complex bodies, lysosomes and nuclei were seen in the cytoplasm. Group B: Secretory cells, high-columnar shaped, increased significantly. In cytoplasm, free ribosomes increased greatly; rough surfaced endoplasmic reticula, mitochondria, Golgi complex bodies were plenty; lysosome decreased; nucleus enlarged; a couple of plasmosomes were seen; the proportion of the nucleoplasm ascended. Group C: Secretory cells were not observed. The edges of smooth muscular cells were clear. In the cytoplasm, mitochondria and rough surface endoplasmic reticula were seen; lipid droplets were rarely seen. The nucleus was normally shaped and the nucleolus was obvious. Group D: Glandular cells were not observed. Edges of smooth muscular cells were clearly seen. In the cytoplasm, mitochondria were observed; endoplasmic reticula were seen to expand; lysosomes increased; many vacuoles were seen. The shape of the nucleus was normal and the plasmosomes were obvious.Conclusion: 1. In group C and D, Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) had obvious removal effects on endometrium of the experimental rat. In Group C, the removal effects were satisfied and there was no big influence to mesenchyma and smooth muscular cells, small dosage was therefore believed as the ideal one. 2. Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) exerted the removal effects on endometrium of experimental rats through mechanisms of decomposing and killing and damaging proliferated cells, thus to make necrosis and scaling of endometrial cells come true. 3. Diaomo Zhibengye (Endometrium-removing and Bleeding-stopping Solution) had a significant targeting function to endometrium of experimental rats, which is expected to be a new drug to treat functional uterine bleeding.
