Objective To detect the expression of neutrophil gelatinase-associated lipocalin (NGAL/Lcn2) in helicobacter pylori (Hp) positive gastric mucous and noninfected. In order to explore the relationship and clinical significance of NGAL/Lcn2,Hp and gastric carcinoma(GC).Method Eight cases of human gastric cacinoma specimens were collected from patients who underwent surgical treatment. Reverse Transcription-Polymerase Chain Reaction(PT-PCR) was used to measure the expression of NGAL/Lcn2 mRNA; Warthin Starry silver staining and Giemsa staining were used to detect the Hp.SP immunohistochemical(IHC) method was employed for detecting the expression of NGAL/Lcn2 in different stages of pre-neoplastic leading to GC.Results1.The relative express quantity of NGAL mRNA in GC (1.025±0.095) was higher than normal (0.689±0.039) and dyplasia gastric mucous tissues (0.895±0.022).It showed with higher frequency in GC.(P≤0.05).2.IHC staining results showed that positive NGAL/Lcn2 expression in the gastric tissues was 65.56%(59/90). It showed NGAL/Lcn2 expression in normal gastric mocus(27.27%),IM(41.67 %), dysplasia(62.96%), early gastric cacinoma (70.00%)and GC (90.00%). In non-neoplastic (n=50) and neoplastic tissue sections(n=40), NGAL/Lcn2 staining was seen with higher frequency.(P≤0.05).In GC, NGAL/Lcn2 staining was seen with similar frequency in differentiation degree.(P>0.05)3.The whole gastric tissues,56 cases of Hp positive were more frequently seen NGAL/Lcn2 than 34 cases of noninfected (P≤0.05).Conclusion1 The high expression of NGAL mRNA in GC, revealing the aberrant expression of NGAL/Lcn2 may play a role in the development of GC.2 NGAL/Lcn2 was more frequently seen in Hp positive cases than in noninfected cases, suggests the overexpression of NGAL in relation to Hp, they may have a causal relation to the development of GC.3 The overexpression of NGAL/Lcn2 may be correlated with early diagnosis of GC and as a useful biornarker for its biological behavior.
