Objective:To probe the Mechanism of COX-2 Regulates E-cadherin by Akt, NF-κB in human gastric carcinoma cells.Methods:After the treatment of celecoxib and PGE2 in different concentration at different time points on human gastric carcinoma cell lines SGC-7901, the MTT method was employed to detect survival rate of the cells. The real-time quantitative PCR was used to detect the level of COX-2,NF-κB,Akt and E-cadherin mRNA respectively. And Western blot was used to detect the corresponding level of COX-2,NF-κB,Akt and E-cadherin protein.Results:①With the intervention of Celecoxib,the mRNA and protein expression of both COX-2 and NF-κB decreased significantly and performed a dose-time dependent manner The Akt mRNA expression has no significant change and the p-Akt protein expression decreased significantly and performed a dose-time dependent manner. While the mRNA and protein expression of E-cadherin increased significantly and performed a dose-time dependent manner.②With the intervention of PGE2, the Akt mRNA expression has no significant change.The p-Akt protein expression increased significantly and performed a dose-time dependent manner. The mRNA expression of NF-κB increased significantly and performed a dose-time dependent manner, while the mRNA and protein expression of E-cadherin declined significantly and performed a dose-time dependent manner.Conclusions:COX-2 Regulates E-cadherin by Akt, NF-κB in human gastric carcinoma cells.
