Objective:To observe the effects of liucha on Lipid metabolism in obese SD rats.Detection the insulin, leptin,adiponectin levels in serum, and adiponectin, adiponectin receptors and its signal transduction pathway related factors in adipose tissue, to explore its mechanism..Methods:Built the obese model by fed high fat diet,use random number table divided rats into 6 groups:Noramal control group, Obese model group, Positive group, Liucha low group, Liucha medium group, Liucha high group, n= 10. Liucha was administered intragastrically to obese rats for eight weeks. Then, body weight, body Length,obese indexes(Lee's), fat weight around kidneys and testis were detected. Automatic biochemical analyzer detected blood glucose and lipid. Producted the tissue sections, then, Observed the quantity and morphology of fat cells. Serum insulin, leptin, adiponectin were measured in each group by Radioimmunoassay and ELISA. Using Real-time quantitative RT-PCR to detect adipose tissue adiponectin, adipoR2, AMPK,PPARa and PPARy mRNA levels.Results:Body weight, Obese indexes(Lee's), fat weight around kidneys and testis of Liucha groups were significantly lower than those in obese model group, while, Under the microscope of 400 times the fat cells diameter were reduceed, the number were increased in the whole field of vision atfet using liucha for 8w. The levels of serum adiponectin of Liucha groups were increased significantly than those in obese model group (P< 0.01), Liucha medium group are best. Adipose tissue adiponectin, adipoR2, AMPK, PPARa and PPARy mRNA and AMPK protein levels of Liucha medium group were increased.Conclusion:Liucha can significantly inhibit rats weight gain which fed high fat diet, improved rats body lipid metabolism. The mechanism may be through regulation the adiponectin signal transduction pathway related genes such as PPAR, AMPK,mRNA and protein levels.To improve insulin resistance in obese rats may be also qne of the mechanisms. In addition, Liucha seems to be not improvement leptin resistance and reduce the role of dietary intake.
