Objective:This study is to investigate the ischemic reperfusion injury with CO2 influence in small pig situ renal transplantation and the expression of Bcl-2 and Bax in renal allografts.Methods:1. Situ retroperitoneal allogeneic renal transplantation model was established in small pig, end-to-end anastomosis was performed between the donor's and the receptor's renal artery, vein and ureter.2.36 matched small pigs which had been tested in blood were randomly divided into group A and B, group A was performed with CO2,group B was performed without CO2. Postoperative serum creatinine value of receptor was detected regularly after the renal transplantation was successful.Pathological were observed at the 3rd,7th,30th day post-transplantation. The immunohistochemistry was used to comprehend localization and expression of Bcl-2 and Bax in allografts.Results:1.Small pig situ retroperitoneal renal transplantation model was established successfully, the survival rate of renal allograft was 80 percent.2.1mmunohistochemistry revealed that Bcl-2 and Bax protein were mainly expressed in the cytoplasm of renal tubules. At the 3rd and 7th day, the expression of Bcl-2 in grafted kidney was significantly lower in CO2 group than that in without CO2 group, but the expression of Bax in allograft was significantly higher in CO2 group than that in without CO2 group (P<0.05). At the 30th day, Both the expression of Bcl-2 and Bax were not significantly different in two groups (P>0.05). Conclusion:1.Small pig situ retroperitoneal renal transplantation model is stable and reliable, the operative procedure is convenient with less complication and high survival rate of allograft, it can be available for the research of ischemic reperfusion injury with CO2 influence in small pig renal transplantation.2.CO2 can promote the procedure of apoptosis and aggravate ischemic reperfusion injury of allograft in the early stage of post-transplantation,maybe by down-regulating the expression of Bcl-2 and by up-regulating the expression of Bax.
