Objective:To study the expression of Sphk-1 and IL-1βin normal brain tissues and human gliomas,and analyze the relationship between the expression of both and the malignancy of gliomas,and the relationship between both of them.To investigate the possible of choosing Sphk-1 and IL-1βto be a target of curing gliomas.Methods In the experiment,62 cases of gliomas were obtained from Nuerosurgery Department of First Hospital of Jilin University during the year of 2008 and 2010. All samples were examined by pathologists and confirmed as primary gliomas so as to be the test group.Meantime,10 cases of normal brain tissues,which were obtained from the patients of cerebral hemorrhage, were to be the control group. To examine the expression of Sphk-1 and IL-1βin gliomas of different gradings and normal brain tissues.And chooseχ2 test and spearman'test to analyze all of the data so as to analyze the correlations between the grading and both.Results In 10 normal brain tissues and 62 gliomas,the positive expression of SphK-1 was 0% and 66.1%,and the positive expression of IL-1βwas 50% and 87.1%.Statistics analysis indicated that the expression of SphK-1 and IL-1βwas much higher than that in mormal brain tissues,which showed significant difference (P<0.05)。In 28 cases of lower-grade gliomas(WHOⅠ-Ⅱ) and 34 case of higher-grade gliomas (WHOⅢ-Ⅳ),the positive expression of SphK-1 was 39.3% and 88.2%, the positive expression of IL-1βwas 89.3% and 85.3%.the expression of SphK-1 in higher-grade gliomas was much higher than in lower-grade gliomas,which showed significant difference (P<0.01)。Though the expression of IL-1βin higher-grade and lower-grade gliomas has significant difference (P<0.05),the difference is negative;In 4 pathologic grades,the positive expression of SphK-1 was gradeⅠ,Ⅱ39.3%(11/28), gradeⅢ81.8%(18/22), gradeⅣ100%(12/12), the positive expression of IL-1βwas gradeⅠ,Ⅱ89.3%(25/28), gradeⅢ77.3%(17/22), grade IV 100 %(12/12), respectively. Throughχ2 test,the expression of SphK-1 was increasing with the grades increasing,which have significant difference (P<0.05).Though IL-1βhas significant difference in different grades glomas (P<0.05),it was negative correlations between grade I-II and III.So the expression of IL-1(3 wasn't considered to be increasing with the grades Increasing. In 10 normal brain tissues and 62 gliomas,the positive expression of SphK-1 and IL-1βwas 40 cases,the SphK-1 positive and IL-1βnegative was 1 cases, the SphK-1 negative and IL-1(3 positive is 19 cases,and both of them which were negative was 12 cases.It indicated that the difference was significant through spearman's andχ2 test.So the expression of them might be considered to have significant correlation.Conclusion:The expresion of SphK-1 in human brain gliomas was significant,which has significant interala-tion with the malignance of gliomas. And meantime,the expression of SphK-1 and IL-1βwere found to have significant correlations.considering the actions which were taken in the malignance and prognosis of SphK-1 and IL-1β, both could be selected to be the target of gene and immunization therapy.
