The interferons represent proteins with antiviral activity that are secreted from cells in response to a variety of stimuli. Interferon can induce cell to produce some proteins which have activities like enzyme. By those proteins interferon can restrain virus coping and damage virus. Interferon also has antitumor activities and immune-regulated function. In general, exposure of cells to viruses and double-stranded RNAs induce the production of IFN-αand IFN-βspecies. Nevertheless, the ratio of these interferons produced by cells varies with the tissue of origin and the species of the organism. Most of the interferons in therapeutic use are produced through genetic engineering. As the first biotherapeutic approved, IFN-αpaved the way for the many new biotherapeutics to follow.In this study, a recombinant IRES-neo-IFN plasmid was digested by PvuI, then the objective gene was recovered and purified. The gene was introduced into male pronucleus of KM mouse fertilized ovum by microinjection, it amounts to 864 fertilized ovum, the survial is about 823, they were transplanted into ampulla of uterine tube of 33 recipient mice. We got 31 mice which have been injected the Human Interferon-αgene, in which 4 was positive by PCR and Southern blotting; F0 transgenic mice are breed with normal mice.All transgenic mice were tested with gene integration by PCR. The result showed that the contract integration rate was about 49.3%(35/71)in the first generation(F1). To analyze the expression pattern of the interert gene in IFN-αtransgenic mice, human IFN-αELISA Kit were used. Serum of four lines F1 generation mice which were positive by PCR identification was collected, ELISA showed the expression of Human Interferon-αin 3 of them. The research indicated that IRES-neo-IFN expression vector not only can express target gene in the original generation transgenic mice and its offspring,but also can be steadily inherited to the posterity of transgenic mice. The successfully establishing of IFN-αtransgenic mice is essential for the study of virus-resistant effect which was induced by IFN-αand may serve as prototypes to generate disease-resistant animals.
