Radix et Rhizoma Salviae red-rooted salvia Miltiorrhiza (lamiaceae),the traditional Chinese medicine with extensive pharmacological activity, is increasingly used with years. The Salvia miltiorrhiza salvia miltiorrhiza polysaccharides (SMP), a kind of natural high polymers, can adjust the immune system, resist oxidation and senescence and exhibite tremendous development potential.Many studies of the chemical ingredients of salvia miltiorrhiza had been reported. But few attention is not paid to the research of polysaccharides, especially the influence factors of the polysaccharide extract. In this study, we focus on the factors affecting the extraction of roots water-soluble polysaccharide from salvia miltiorrhiza and then determine the optimum extraction process parameters. The monosaccharides, average molecular weight andbiological characteristics of the polysaccharide were analysed. By the antitumor and hypoxia tolerance test of salvia miltiorrhiza polysaccharide in mice, the amount of anti-tumor effect relation was determined. These results p rovides theory basis for the further development and utilization of the polysaccharides salvia miltiorrhiza. The main results are as follows:1. According to the water extraction of polysaccharides from Salviamiltiorrhiza, the optimum weight ratio of water and HDP is 1:20. The optimum extracting time is 4 hours. Theoptimum ethanol concentration is90%. The optimum parameters for the ultrasonic is salvia miltiorrhiza weight and water ratio of 1:20, extracting time of 40 minutes, ethanol concentration of 90%.2. Whether white or purple flower salvia miltiorrhiza in HDP, the most abundant polysaccharide is in leaves and the next is in root and stem. During the same harvest time, the acquisition of salvia miltiorrhiza polysaccharide is different in different subspecies. The salvia miltiorrhizawith white flower contains more polysaccharide than that of purple flower. Upon the different harvest periods, acquisition roots polysaccharide inoctober is higher than that in may. 3. Two homogeneous components were obtained from the polysaccharide out of the free proteins, the average molecular weight of which are 1.389×10~6,4.028×10~5 Da by HPLC. The main components of monosaccharide of the polymer are glucose, galacose arabinose and rhamnose.4. The significant difference macrophage phagocytosis rate and index (P < 0.05) betweenthe high dose group and control shows the SMP had obviously increased the abdominal macrophage phagocytosis rate andindex and the non-specific immune function of normal mice can be improved. The significant difference of spleen and thymic index between theCyclophosphamide + high-dosage groups and individual cyclophosphamide (P < 0.05) indicated the high doses of salvia miltiorrhiza polysaccharides can significantly improved cyclophosphamide mice immune function. High doses of SMP can stimulate the transformation of T lymphocytes, improve the appreciation ability of T lymphocytes. This indicated that polysaccharide can enhance the host immune function so as to inhibit the growth of tumor cells, and be free from side effects. So the salvia miltiorrhiza polysaccharides is an ideal anti-tumor medicine.5.Different doses of SMP can prolong the survival time under the deficiency of oxygen in different levels. The survival time of the 100 to 400ml/kg doses groups were significantly longer than that of controls (P < 0.01), of which the 200ml/kg is the optimum dose. 6, The weight loading swimming duration directly surpported that salvia miltiorrhiza polysaccharide can prolong the swimming time of mice because it can improve the body's energy metabolism and reduce the protein and nitrogen catabolism.This can reduce blood ureanitrogen and then show anti-fatigue effect.
